-The banana crop stands out as an activity of great social and economic importance in Brazil, which occupies the fifth place in world production. Synthetic seed production is becoming promising for a micropropagation and in vitro conservation. The aim of the study was to analyze the conversion and growth of 'Prata-anã' banana's microshoots clone Gorutuba from synthetic seed in MS medium and vermiculite, different substrates and concentrations of BAP (6-benzylaminopurine) associated with ANA (acetic naphthalene acid) in the constitution of its capsule were tested. The microshoots were immersed in the sodium alginate matrix (3%) and dripped in a solution of CaCl 2 .2H 2 O (100 mM) for complexation and then in KNO 3 solution (100 mM) to decomplex. The experimental design was completely randomized in a 2 x 5 factorial design (substrate x BAP concentrations), containing different substrates (MS culture medium and vermiculite) and BAP concentrations (2.22, 4.44, 6.66, 8.88 and 13.32 μmol L -1 ) associated with NAA (naphthalene acetic acid) 0.54 μmol L -1 , totaling 10 treatments, with 4 replicates, and that each replicate containing 5 seeds. The evaluations of conversion, number of leaves, leaf length, leaf height, number of roots, root length and oxidation were performed at 30 and 60 days.The use of the MS medium provided better growth results in relation to vermiculite as substrate, in which the different BAP concentrations did not differ from each other. It was found that, in MS culture medium, BAP concentrations above 8.88 μmol L -1 in the capsule composition are not indicated for microshoots growth. This work was conducted with the objective of evaluating the performance of cactus pear cultivars on in vitro culture using bioreactors and the conventional method. The treatments were arranged in a 3 × 2 factorial scheme with a completely randomized design, and the three cactus pear cultivars (Orelha de elefante mexicana, Miúda and IPA-Sertânia) were combined with two micropropagation methods (conventional and temporary immersion bioreactors), totaling six treatments with five replicates each. After 30 days, the following variables were evaluated: cladode length, fresh explant matter mass, number of shoots and number of roots. There was a significant interaction between the cultivar factor and the micropropagation method for the cladode length and fresh matter mass, with bioreactors being the most responsive. However, regarding to the number of shoots and number of roots, there was no interaction between these factors under the same level of significance, where the conventional crop stood out. The results indicate different morphogenetic responses among the tested cultivars, and specific in vitro propagation protocols should be developed for each one.
Modification of in vitro culture environment can improve the development of plants, obtaining higher morphological and physiological quality. The objective of this work was to analyze the influence of different luminous spectrums as well as different doses of auxin NAA on in vitro development of cactus pear (Opuntia ficus-indica (L.) Mill). The experiment was conducted in a completely randomized design in factorial scheme 4 x 3 (Luminous spectrums x NAA doses), corresponding to four luminous spectrums: red, green, blue and white, and three doses of NAA 1, 2 and 3 mg. L-1. A total of 12 treatments with 6 replications were applied including 1 plant per repetition. Cactus pear plants were introduced on MS culture medium solidified with 7% agar, and pH adjusted to 5.8. We observed higher development of plants (growth and rooting) under luminous spectrums, 30 and 60 days after plant establishment (red and green). At concentrations of 1 and 2 mg L-1 of NAA we observed better height of the plants, 30 and 60 days after establishment. The luminous spectrums red and green influenced the in vitro development of cactus pear plants. The best doses to promote better plant growth are 1 and 2 mg. L-1 of NAA.
