Phonon-cavity electromechanics allows the manipulation of mechanical oscillations similar to photon-cavity systems. Many advances on this subject have been achieved in various materials. In addition, the coherent phonon transfer (phonon Rabi oscillations) between the phonon cavity mode and another oscillation mode has attracted many interest in nanoscience. Here, we demonstrate coherent phonon transfer in a carbon nanotube phonon-cavity system with two mechanical modes exhibiting strong dynamical coupling. The gate-tunable phonon oscillation modes are manipulated and detected by extending the red-detuned pump idea of photonic cavity electromechanics. The first- and second-order coherent phonon transfers are observed with Rabi frequencies 591 and 125 kHz, respectively. The frequency quality factor product fQ ∼ 2 × 10 Hz achieved here is larger than kT/h, which may enable the future realization of Rabi oscillations in the quantum regime.
The formation of α-synuclein aggregates is a major pathological hallmark of Parkinson's disease. Copper promotes α-synuclein aggregation and toxicity in vitro . The level of copper and copper transporter 1, which is the only known high-affinity copper importer in the brain, decreases in the substantia nigra of Parkinson's disease patients. However, the relationship between copper, copper transporter 1 and α-synuclein pathology remains elusive. Here, we aim to decipher the molecular mechanisms of copper and copper transporter 1 underlying Parkinson's disease pathology. We employed yeast and mammalian cell models expressing human α-synuclein, where exogenous copper accelerated intracellular α-synuclein inclusions and silencing copper transporter 1 reduced α-synuclein aggregates in vitro , suggesting that copper transporter 1 might inhibit α-synuclein pathology. To study our hypothesis in vivo , we generated a new transgenic mouse model with copper transporter 1 conditional knocked-out specifically in dopaminergic neuron. Meanwhile, we unilaterally injected adeno-associated viral human-α-synuclein into the substantia nigra of these mice. Importantly, we found that copper transporter 1 deficiency significantly reduced S129-phosphorylation of α-synuclein, prevented dopaminergic neuronal loss, and alleviated motor dysfunction caused by α-synuclein overexpression in vivo . Overall, our data indicated that inhibition of copper transporter 1 alleviated α-synuclein mediated pathologies and provided a novel therapeutic strategy for Parkinson's disease and other synucleinopathies.
The buckling deformation and stress distribution of the tubing nearby the packer would be seriously influenced by the packer constraints. This paper focused on the Strength analysis of transitional segment. The method to calculate the buckling deformation and the equivalent stress of transitional segment were derived considering the tubing boundary and continuity condition, adopting the fourth-order nonlinear ordinary differential equations and helical buckling strength method. This study makes up for the weakness of traditional stress analysis of the buckling tubing nearby the packer in vertical well, improves the pertinence and accuracy of stress analysis of buckling tubing. The results of analysis show that with the axial pressure increasing lead to the length of the transitional segment irregular fluctuations and the maximum equivalent stress increasing. The equivalent stress would gradually reduce and finally approach a constant.
In the deep and ultra deep well casing wear is serious. The decrease of strength leads to casing damage, affects the follow-up well test and completion. In order to analyze the influence of the contact force, rotating speed, steel grade and muds on casing wear, determine the optimized level collocation scheme which makes casing wear minimum, optimize drilling parameters and casing programs, reduce casing wear, four factors and three levels orthogonal experiment of casing wear was carried out, and the range analysis of the experiment results is made. The results show that the best match of factors is A1B1C1D3, namely when the contact force is 200 N(A1), rotating speed 60 r/min (B1), steel grade N80(C1), oil-based mud (D3) casing wear is minimal. The influence of muds on casing wear is the largest, the next is casing steel grade, and then rotating speed, the effect of the contact force minimal. The improvement of mud performance is best method to reduce casing wear.
