In rice (Oryza sativa), the presence of a dominant Badh2 allele encoding betaine aldehyde dehydrogenase (BADH2) inhibits the synthesis of 2-acetyl-1-pyrroline (2AP), a potent flavor component in rice fragrance. By contrast, its two recessive alleles, badh2-E2 and badh2-E7, induce 2AP formation. Badh2 was found to be transcribed in all tissues tested except for roots, and the transcript was detected at higher abundance in young, healthy leaves than in other tissues. Multiple Badh2 transcript lengths were detected, and the complete, full-length Badh2 transcript was much less abundant than partial Badh2 transcripts. 2AP levels were significantly reduced in cauliflower mosaic virus 35S-driven transgenic lines expressing the complete, but not the partial, Badh2 coding sequences. In accordance, the intact, full-length BADH2 protein (503 residues) appeared exclusively in nonfragrant transgenic lines and rice varieties. These results indicate that the full-length BADH2 protein encoded by Badh2 renders rice nonfragrant by inhibiting 2AP biosynthesis. The BADH2 enzyme was predicted to contain three domains: NAD binding, substrate binding, and oligomerization domains. BADH2 was distributed throughout the cytoplasm, where it is predicted to catalyze the oxidization of betaine aldehyde, 4-aminobutyraldehyde (ABald), and 3-aminopropionaldehyde. The presence of null badh2 alleles resulted in AB-ald accumulation and enhanced 2AP biosynthesis. In summary, these data support the hypothesis that BADH2 inhibits 2AP biosynthesis by exhausting AB-ald, a presumed 2AP precursor.
Antimicrobial resistance against colistin has emerged worldwide and is threatening the efficacy of colistin treatment of multi-resistant Gram-negative bacteria. In this study, PCRs were used to detect mcr genes (mcr-1, mcr-2, mcr-3) in 213 anal and 1,339 nasal swabs from pigs (n = 1,454) in nine provinces of China, and 1,696 cloacal and 1,647 oropharyngeal samples from poultry (n = 1,836) at live-bird markets in 24 provinces. The mcr-1 prevalences in pigs (79.2%) and geese (71.7%) were significantly higher than in chickens (31.8%), ducks (34.6%) and pigeons (13.1%). The mcr-2 prevalence in pigs was 56.3%, significantly higher than in chickens (5.5%), ducks (2.3%), geese (5.5%) and pigeons (0%). The mcr-3 prevalences in pigs (18.7%), ducks (13.8%) and geese (11.9%) were significantly higher than in chickens (5.2%) and pigeons (5.1%). In total, 173 pigs and three chickens were positive for all three mcr genes. The prevalences of the mcr were significantly higher in nasal/oropharyngeal swabs than in the anal /cloacal swabs. Phylogenetic studies identified 33 new mcr-2 variants and 12 new mcr-3 variants. This study demonstrates high prevalences of mcr in pigs and poultry in China, and indicates there is need for more thorough surveillance and control programs to prevent further selection of colistin resistance.
The main objective of this study was to investigate the individual cow effect of bovine leukemia virus (BLV) infection on milk production and somatic cell score (SCS). The fluorescence resonance energy transfer (FRET) quantitative PCR established in this study and a commercial ELISA kit revealed that 49.1% of dairy cattle (964/1,963) from 6 provinces of China and 1.6% of beef cattle (22/1,390) from 15 provinces were BLV positive. In a detailed study of 105 cows, BLV was found most commonly in buffy coat samples that also had highest copy numbers (10(4.75±1.56) per mL); all cows negative for BLV in buffy coat samples were also negative in vaginal swab, milk, and fecal samples. Copy numbers of BLV were 10(2.90±0.42)/gram of feces, 10(0.83±0.62)/mL of milk, and 10(2.18±0.81) per vaginal swab. The BLV-positive cows had significantly lower milk production in the early (26.8 vs. 30.9kg) and middle stages of lactation (22.2 vs. 26.1kg) in animals with ≥4 parities than the BLV-negative cows; they also had significantly higher SCS in early and middle lactation stages (early=5.2 vs. 4.3; middle=4.9 vs. 3.9) in animals with ≥4 parities. Milk production and SCS did not significantly differ between the BLV-infected and -uninfected cows when they were in the late lactation stage or in animals with ≤3 parities. Taken together, our results indicate that BLV infections are widespread in the dairy farms of China. Vaginal secretions and feces may be involved in BLV transmission. A BLV infection may result in reduced milk yield and increased SCS in a parity and lactation stage-restricted manner.
