15N2 measurement of nitrogen fixation by legumes and actinorhizals: theory and practice

Abstract: A gas-tight chamber has been constructed to calibrate the tSN isotope dilution method against direct L5N 2 measurements. The theoretical basis for such estimates is given, and the practical problems associated with the experiments are discussed.

“…We determined d15N in shoots of legume and non-legume material collected in June 1995, October 1995, June 1996, and October 1996Finnigan MAT, Bremen, Germany). The fraction of legume N originating from symbiotic fixation was calculated as %N dfa ¼ ( 15 N nonl À 15 N leg )/( 15 N nonl À 15 N atm ), where 15 N leg , 15 N nonl , and 15 N atm are the 15 N mole fractions of N in legumes, all non-legume plants present in the plot, and atmospheric N 2 , respectively (Wood and McNeill 1993). Plant N and P pools were determined by dry combustion (CHN-900, LECO Instruments) and by a micro-Kjehldahl-type wet digestion and colorimetric determination of PO 4 3À (Niklaus et al 1998a).…”

“…where 15 N leg , 15 N nonleg , and 15 N atm are the 15 N-mole fractions of the total N in legumes, all nonlegume plants present in the plot, and atmospheric N 2 , respectively (Wood and McNeill 1993). Ecosystem-level nitrogen retention was estimated as the amount of the 15 N label applied in 1995 that was recovered either in the plant biomass harvested in subsequent years, or still remained in plants, microbes, litter, or soil at termination of the experiment five years later.…”
