2-Deoxyglucose combined with wild-type p53 overexpression enhances cytotoxicity in human prostate cancer cells via oxidative stress

Abstract: Over expression of the tumor suppressor gene, wild type p53 (wtp53), using adenoviral vectors (Adp53) has been suggested to kill cancer cells by hydroperoxide-mediated oxidative stress [1,2] and nutrient distress induced by the glucose analog, 2-deoxyglucose (2DG), has been suggested to enhance tumor cell killing by agents that induce oxidative stress via disrupting hydroperoxide metabolism [3,4]. In the current study clonogenic cell killing of PC-3 and DU-145 human prostate cancer cells (lacking functional p5… Show more

“…Replication-incompetent adenoviral vectors, Ad-empty (AdE), 25 MOI AdCMV catalase (AdCat), 25 MOI AdCMV glutathione peroxidase 1 (AdGPx1), 50 MOI AdCMV SOD1 (AdSOD1), and 50 MOI AdCMV SOD2 (AdSOD2) vectors are previously described (Zwacka et al, 1998; Ahmad et al, 2005, 2008). 20 MOI AdFerritin heavy and light chain (AdFtH and AdFtL, respectively) were generated by ViraQuest (North Liberty, IA) using commercially available iron regulatory element (IRE)-lacking vectors (OriGene).…”

“…In the human cancer cell lines tested in the present study, only MCF-7 cells express wild-type p53 protein, while other cell lines lack the functional p53 protein [41][42][43]. Since p53 is activated by treatment with resveratrol [39], it is possible that its activation may mask the growth-stimulatory effect in cells that express the wild-type p53 protein.…”

“…The glucose analog, 2DG, is a clinically relevant inhibitor of glucose metabolism believed to mimic the effects of glucose deprivation [20][21][22]. Therefore, we hypothesized that applying 2DG and DOX and modulating intracellular thiol levels using BSO will inhibit critical aspects of thiol-mediated hydroperoxide metabolism.…”
