20(S)-Protopanaxadiol-Induced Apoptosis in MCF-7 Breast Cancer Cell Line through the Inhibition of PI3K/AKT/mTOR Signaling Pathway

Abstract: 20(S)-Protopanaxadiol (PPD) is one of the major active metabolites of ginseng. It has been reported that 20(S)-PPD shows a broad spectrum of antitumor effects. Our research study aims were to investigate whether apoptosis of human breast cancer MCF-7 cells could be induced by 20(S)-PPD by targeting the Phosphatidylinositol 3-kinase/Protein kinase B/Mammalian target of rapamycin (PI3K/AKT/mTOR) signal pathway in vitro and in vivo. Cell cycle analysis was performed by Propidium Iodide (PI) staining. To overexpre… Show more

“…The PI3K/Akt/mTOR signaling pathway has been reported to exert vital function in cell proliferation, differentiation, migration and invasion [31,32]. In our present study, we explored the correlation between SNHG20 and PI3K/Akt/mTOR signaling.…”

“…This pathway serves a vital role in regulating cell proliferation, apoptosis, differentiation and metabolism by affecting the activation state of several downstream effector molecules (2628). In recent years, abnormal activation of the PI3K/Akt/mTOR pathway has been detected in numerous human malignant tumors, and activation of this pathway may be a critical factor leading to the proliferation and metastasis of tumor cells (2931).…”

“…Hence, the expression levels of p-PI3K (p85) [ 32 ], p-AKT (Ser473) [ 33 ], t-PI3K, t-AKT, and p-mTOR (Ser2448) [ 33 ] were investigated after 24 h treatment with AAP-H. As shown in Figure 3 , AAP-H treatment led to a dose-dependent reduction in the levels of p-AKT (Ser473), p-PI3K (p85), and p-mTOR (Ser2448), whereas t-AKT and t-PI3K levels remained unaltered compared to the nontreated group. Multiple signaling pathways operate and cross-talk inside cells; hence, when one pathway is stimulated, the levels of proteins related to that pathway, as well as other associated pathways, change concomitantly [ 34 ].…”

“…The expression of members of the PI3K/AKT/mTOR signaling pathway (p-AKT (Ser473), p-PI3K (p85) and p-mTOR (Ser2448)) in tumor tissue from nude mice was examined using immunocytochemistry assay [ 33 ]. As shown in Figure 7 , p-AKT (Ser473), p-PI3K (p85), and p-mTOR (Ser2448) were detected in the cytoplasm and nucleus of the tumors; however, no expression was observed in the negative control group, whereas strong expression was observed in the positive control group.…”
