2017 White Paper on Recent Issues in Bioanalysis: Rise of Hybrid LBA/LCMS Immunogenicity Assays (Part 2: Hybrid LBA/LCMS Biotherapeutics, Biomarkers & Immunogenicity Assays and Regulatory Agencies’ Inputs)

Abstract: The 2017 11th Workshop on Recent Issues in Bioanalysis (11th WRIB) took place in Los Angeles/Universal City, California on 3–7 April 2017 with participation of close to 750 professionals from pharmaceutical/biopharmaceutical companies, biotechnology companies, contract research organizations and regulatory agencies worldwide. WRIB was once again a 5-day, weeklong event – a full immersion week of bioanalysis, biomarkers and immunogenicity. As usual, it was specifically designed to facilitate sharing, reviewing,… Show more

“…LBA and IC-LC/MS assay performance was evaluated based on fit-for-purpose approach with limited precision, accuracy and specificity as discussed previously [28]. Briefly, the LBA-based assay standard curve was fit using a 4-parameter logistic regression (4PL) algorithm with 1/y weighted regression using SoftMax Pro 7 Software (Molecular Devices, San Jose, CA).…”

“…Selection of suitable surrogate peptides from either light chain or heavy chain with good sensitivity and high specificity is critical to the quantitative accuracy and reliability of the assay. It is important to carefully evaluate both immuno-capture and proteolytic digestion efficiencies in order to develop a robust and sensitive method [95,96]. Stable isotopically labeled internal standards (SIL-IS) are usually incorporated as early in the workflow as possible to normalize as much of the process as possible for improved accuracy and precision of quantification.…”
