3D Scaffolds of Polycaprolactone/Copper-Doped Bioactive Glass: Architecture Engineering with Additive Manufacturing and Cellular Assessments in a Coculture of Bone Marrow Stem Cells and Endothelial Cells

Abstract: The local delivery of Cu 2+ from copper-doped bioactive glass (Cu-BaG) was combined with 3D printing of polycaprolactone (PCL) scaffolds for its potent angiogenic effect in bone tissue engineering. PCL and Cu-BaG were, respectively, dissolved and dispersed in acetone to formulate a moderately homogeneous ink. The PCL/Cu-BaG scaffolds were fabricated via direct ink writing into a cold ethanol bath. The architecture of the printed scaffolds, including strut diameter, strut spacing, and porosity, were investigate… Show more

“…Bone marrow samples were obtained with the donor’s written informed consent and processed under ethical approval of the Ethics Committee of the Expert Responsibility area of Tampere University Hospital, Tampere, Finland (R15174). The cells were isolated as described previously ( Wang et al, 2019 ). The BMSCs were cultured in α -modified minimum essential medium ( α -MEM; Gibco) supplemented with 5% human serum (HS; BioWest or Serana), 100 U/ml penicillin, 100 µg/ml streptomycin (Pen/Strep; Lonza), and 5 ng/ml basic fibroblast growth factor (bFGF; Miltenyi Biotec) and used between passages 3 and 6.…”

“…Moreover, Cu ions were shown to promote wound healing in rats, which has been linked to the up regulation of vascular endothelial growth factor (VEGF) by stimulated cells [6] and to the antimicrobial behavior of Cu. Indeed, Cu is able to artificially mimic hypoxia, which plays an important role in blood vessel formation as well as in the differentiation and recruitment of endothelial cells [37,39]. The angiogenic effect of Cu-doped 45S5 BG has been investigated and proven in vitro and in vivo using 3D BG scaffolds [40].…”

“…The HUVECs were cultured at +37 °C in 5% CO 2 , and the medium was changed twice per week. To verify the endothelial phenotype of HUVECs, surface marker expression was characterized by flow cytometry (FACSAria; BD Biosciences, Erembodegem, Belgium) as described previously [ 30 ].…”
