5‐Hydroxytryptamine (5‐HT)‐exacerbated DSS‐induced colitis is associated with elevated NADPH oxidase expression in the colon

Abstract: Aim: This study investigated the impact of 5-hydroxytryptamine (5-HT) on the expression of NOXs in dextran sulfate sodium (DSS)-induced colitis in mice. Methods: C57BL/6J (B6) mice at 6 to 8 weeks of age were treated with 5-HT, DSS, or DSS + 5-HT. After 6-day treatment, the severity of colitis, infiltration of leukocytes, and messenger RNA (mRNA) and/or protein levels of Nox1, Nox2, Nox4, and Duox2 were analyzed in the colon by real-time quantitative polymerase chain reaction (qPCR), immunohistochemistry (IHC)… Show more

“…For immunohistochemical (IHC) staining, mouse anti-rabbit F4/80 (GB11027, Servicebio, Hubei, China, 1:300 dilution) was used as the primary antibody, and goat anti-rabbit IgG was the secondary antibody (GB23303, Servicebio, Hubei, China, 1:200 dilution). IHC was performed using previously described protocols (Dong et al, 2019). Negative control sections were treated in the same manner, with omission of the primary antibody.…”

“…[ 146 ] On the contrary, mice treated with 1% DSS or DSS + 5‐HT showed an exacerbation in colitis that further increased NOX expression in mice colons. [ 183 ] In the mouse model, spontaneous chronic colitis is associated with an increase in 5‐HT levels. [ 184 ] However, new clues suggest that 5‐HT modulate the intestinal microbiota composition by acting on 5‐HTRs exert pro/anti‐inflammation, [ 31,162,185 ] and upregulation of the pro‐inflammatory cytokines (IL‐6, TNF‐α, and IL‐8) through 5‐HT binding to its receptors, namely 5‐HTR3, 5‐HTR4, and 5‐HTR7 in DCs.…”

“…DSSinduced colitis is associated with elevated NADPH oxidase expression in the colon. 30 Pro-inflammatory cytokines including TNF-α, IL-17 and IL-23 have been considered to play a central role in the processes of UC. 31 PGE2 was increased significantly in patients with IBD, which was important indicator for evaluating the degree of UC.…”
