Bacterial Associates Modify Growth Dynamics of the Dinoflagellate Gymnodinium catenatum

Marine phytoplankton cells grow in close association with a complex microbial associate community known to affect the growth, behavior, and physiology of the algal host. The relative scale and importance these effects compared to other major factors governing algal cell growth remain unclear. Using algal-bacteria co-culture models based on the toxic dinoflagellate Gymnodinium catenatum, we tested the hypothesis that associate bacteria exert an independent effect on host algal cell growth. Batch co-cultures of G. catenatum were grown under identical environmental conditions with simplified bacterial communities composed of one-, two-, or three-bacterial associates. Modification of the associate community membership and complexity induced up to four-fold changes in dinoflagellate growth rate, equivalent to the effect of a 5°C change in temperature or an almost six-fold change in light intensity (20–115 moles photons PAR m-2 s-1). Almost three-fold changes in both stationary phase cell concentration and death rate were also observed. Co-culture with Roseobacter sp. DG874 reduced dinoflagellate exponential growth rate and led to a more rapid death rate compared with mixed associate community controls or co-culture with either Marinobacter sp. DG879, Alcanivorax sp. DG881. In contrast, associate bacteria concentration was positively correlated with dinoflagellate cell concentration during the exponential growth phase, indicating growth was limited by supply of dinoflagellate-derived carbon. Bacterial growth increased rapidly at the onset of declining and stationary phases due to either increasing availability of algal-derived carbon induced by nutrient stress and autolysis, or at mid-log phase in Roseobacter co-cultures potentially due to the onset of bacterial-mediated cell lysis. Co-cultures with the three bacterial associates resulted in dinoflagellate and bacterial growth dynamics very similar to more complex mixed bacterial community controls, suggesting that three-way co-cultures are sufficient to model interaction and growth dynamics of more complex communities. This study demonstrates that algal associate bacteria independently modify the growth of the host cell under non-limiting growth conditions and supports the concept that algal–bacterial interactions are an important structuring mechanism in phytoplankton communities.

The Middle Pleistocene fluvial channel site of the Upper Fossil-bearing Interval at Mata Menge in the So’a Basin, Flores, Indonesia, has yielded the earliest fossil evidence for Homo floresiensis in association with stone artefacts and fossils of highly endemic insular fauna, including Stegodon, giant rats, crocodiles, Komodo dragons, and various birds. A preliminary taphonomic review of the fossil material here aimed to provide additional context for the hominin remains in this bonebed. Analysis was performed on two subsets of material from the same fluvial sandstone layer. Subset 1 comprised all material from two adjacent one-metre square quadrants (n = 91), and Subset 2 all Stegodon long limb bones excavated from the same layer (n = 17). Key analytical parameters included species and skeletal element identification; fossil size measurements and fragmentation; weathering stages; bone fracture characteristics; and other biological and geological bone surface modifications. Analysis of Subset 1 material identified a highly fragmented assemblage with a significant bias towards Stegodon. A large portion of these bones were likely fractured by trampling prior to entering the fluvial channel and were transported away from the death-site, undergoing surface modification causing rounding. Subset 2 material was less likely to have been transported far based on its limited susceptibility to fluvial transport. There was no significant difference in weathering for the long limb bones and fragments, with the highest portion exhibiting Stage 2 weathering, indicating that prior to final burial, all material was exposed to prolonged periods of surface exposure. Approximately 10% of all material have characteristics of fracturing on fresh bone, contributing to the taphonomic context for this bonebed; however insufficient evidence was found for anthropogenic modification.

One of the efforts to improve language skills of the lecturers is by joining IELTS Preparation program organized by one of the International Education institution (IEDUC) located in Bandung. This research aims to find out whether there is a correlation between the results of the Super-IELTS Intensive program (SUPIN) lecturer final score with the real International Test score in sort of the evaluation process on that program. The subject was taken from two classes program comprising 19 lecturers as participants. The data in this study were obtained from the pre and post-test score during SUPIN program and the score on the real International test conducted by the British Council. The result of the research revealed the fact that listening test obtained the highest gain while speaking got the lowest gain during the program. The further analyzing on Variance got a significant linear regression by having 0.959 as a coefficient correlation. It means that there is a significant correlation between final score of SUPIN program (X) and International test score (Y) which contribute 92.1% toward the International test score. Meanwhile, only 7.9% was contributed by other factors. In other words, the SUPIN program is effective to increase Lecturer’s skill on doing IELTS test.
