Abstract: The IRES from poliovirus and from encephalomyocarditis virus (EMCV) added between the cap and the AUG initiator codon were strong inhibitors of chloramphenicol acetyltransferase gene expression in three different cell types. The poliovirus IRES also inhibited bGH (bovine growth hormone) cDNA expression in the HC11 mammary cell line when added between the rabbit whey acidic gene promoter and the cDNA whereas the HTLV-1 IRES showed a stimulatory effect in the same situation. RNA stem loops were added before HTLV… Show more

“…*, P Ͻ 0.05. modules can give rise to artificial IRES activity (40). Furthermore, when the R region of the HTLV-1 5=UTR is attached to other genetic elements, it is known to stimulate gene expression (41,42). To rule out the possibility that the ⌬EMCV sequence together with the HTLV-1 5=UTR could artificially exhibit IRES activity, the luciferase activities from bicistronic reporters containing the ⌬EMCV sequences (dl HTLV-1 5=UTR) or not (dl ⌬⌬EMCV HTLV-1 5=UTR) were measured (Fig.…”

“…This study used full length cDNA sequences for the expression of Cxs, including untranslated regions (UTRs), rather than purely coding sequence. Intercistronic length has been shown to affect IRES-dependent bicistronic expression with greater intercistronic length adversely affecting translation [17]. Both Cx sequences contain large 3' UTRs contributing to intercistronic length that may interfere with IRES-dependent translation.…”
