Abstract: The role of 5-lipoxygenase metabolites of arachidonic acid in the inflammatory response associated with experimental acute pancreatitis has been evaluated. For this purpose, an experimental necrohemorrhagic pancreatitis was induced in rats by intraductal administration of 5% sodium taurocholate. Neutrophil infiltration was detected in pancreas at 1 and 3 h after the induction of pancreatitis. This was concomitant with increased levels of leukotriene B4 and peptide leukotrienes (C4, D4 and E4). In lung, similar… Show more

“…Closa and co-workers demonstrated that xanthine oxidase is involved not only in pancreatic damage but also in the lung inflammation associated with experimental acute pancreatitis [133]. During the early stages of acute pancreatitis, α-amylase is absorbed from ascites fluid and once in the circulation promotes the release of XO by breaking down the binding of XDH/XO to glycoproteins of the endothelial cells.…”

“…There is some evidence that it inhibits the lipoxygenase pathways, thus reducing formation of the leukotrienes, which are proinflammatory autacoids that have also been implicated as mediators of inflammation in acute pancreatitis. 2834 In addition, it has been suggested that diclofenac is much more potent than other NSAID types in its ability to inhibit phospholipase A2, an enzyme strongly implicated in the pathogenesis of acute pancreatitis. These distinct effects of diclofenac may explain the significant efficacy of diclofenac in preventing PEP in this study.…”
