The interactions of bovine serum albumin (BSA) with ionic surfactants (sodium dodecyl sulfate, SDS, and cetyltrimethylammonium bromide, CTAB) and β-cyclodextrin (β-CD) have been investigated by electron paramagnetic resonance (EPR) and circular dichroism measurements. The spin probe selected to report on the interaction of albumin with surfactants and/or β-CD was 4-N,N-dimethyl hexadecyl ammonium-2,2,6,6-tetramethylpiperidine-1-oxyl iodide (CAT16), on account of (a) its balance between electrostatic and hydrophobic character and (b) the ability of BSA to form complexes with various organic molecules. The distribution of the spin probe among different environments in solutions containing only BSA was confirmed by the existence of two components in the EPR spectra: one revealing a restricted mobility of the spin probe, attributed to the protein-spin probe complex, and another one showing free movement, attributed to the spin probe in solution. The presence of surfactants and/or β-CD alters the distribution of CAT16 between various compartments in each system. Formation of protein aggregates as a result of thermal denaturation was evidenced by the appearance of an immobilized component in the EPR spectrum. This component is not present in the EPR spectra of CAT16 in protein/surfactant or protein/cyclodextrin solutions. Circular dichroism spectra of BSA provided information about changes in the secondary structure of the protein induced by the presence of surfactants and/or cyclodextrin in solution. The results demonstrate that β-CD hinders the interaction between the employed surfactants and the protein. The cationic surfactant (CTAB) induces changes in protein conformation at a lower concentration compared to the anionic surfactant (SDS).

Antioxidants play a key role in counteracting the adverse effects of oxidative stress in human organisms. Thus, there is a huge demand for the development of smart and convenient assays for the evaluation of antioxidant activity of synthetic and natural compounds, food samples, plant extracts, etc. The design of a solid-state, flexible, and portable format of the traditional in vitro 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical test is reported. For the first time, purple DPPH radicals are physically immobilized into a silicone matrix by means of a facile and rapid process. The working principle of the resulting sensor is based on the colorimetric process that is associated with the redox reaction of DPPH radicals with antioxidants. When an antioxidant reacts with the sensor, a color change from purple to yellow can be perceived by the naked eye. The response of the sensor is investigated qualitatively and quantitatively toward food samples and selected antioxidants of different natures, and solubilized in different media. It is demonstrated that the DPPH silicone sensor is highly versatile and can be used as a ready-to-use sensor for direct colorimetric detection of antioxidants.

The effect of cationic surfactant Cetyltrimethylammonium bromide (CTAB) molecules on the oxidation of L-threonine by N-bromophthalimide (NBP) has been studied at 308 K. The reaction exhibits first order dependence on NBP and fractional positive order on L-threonine and negative fractional order dependence on HClO 4. The effects of KCl, KBr, phthalimide, mercuric acetate and dielectric constant of the medium have also been studied and summarized. CTAB strongly catalyzes the reaction and after achieving a maximum the rate remains almost constant at higher concentrations of CTAB. This micellar effect has been analyzed, plotted and results are treated quantitatively in terms of Berezin model. The various activation parameters in presence and absence of CTAB have been also evaluated. A suitable mechanism along with rate law consistent with the experimental findings has been worked out. Based on the model association rate constants have been computed.
