A cerebral glioma model for experimental therapy andin vivo invasion studies in syngeneic BD IX rats

Abstract: An in vivo glioma model was developed in syngeneic BD IX rats. The BT4An tumor was derived from serial in vivo passages of the BT4A tumor, originally induced from transformed fetal rat brain cells after transplacental exposure to ethylnitrosourea. The cell line was characterized for the presence of neuroglial differentiation markers, chromosome content and cell cycle distribution as determined by flowcytometry. A standardized method for i.c. tumor induction was developed, and the tumors were investigated by li… Show more

“…The ACBT cells were cultured in DMEM (Gibco, Carlsbad, CA) with high glucose and supplemented with 2 mM L L -glutamine, penicillin (100 U ml )1 ), streptomycin (100 lg ml )1 ), and 10% heat-inactivated fetal bovine serum (Gibco, Carlsbad, CA). The BT 4 C cell line was derived originally from transformed fetal rat brain cells after exposure to ethylnitrosourea [35]. The BT 4 C cells were grown as monolayers in RPMI medium with 10% heatinactivated newborn calf serum (FCS) at 37°C and 5% CO 2 .…”

“…Major hallmarks in GBM in vivo modeling are the glioma models established by Rolf Bjerkvig and colleagues by serial passaging glioma spheroid cultures in nude rats [152,153]. The tumor spheroids keep their genetic profile in culture, as shown by comparative genomic hybridization (CGH) [154].…”

“…A complete description of this model has been provided elsewhere [22]. Briefly, to establish intracranial tumors, the anesthetized rats were fixed in a stereotactic frame (David Kopf Instruments, Tujunga, CA), the skin was incised and a 1.0-mm burr hole was made to fit the following coordinates: 3 mm posterior to and 2 mm to the right of the bregma and depth of 3 mm.…”
