A facile isoelectric focusing of myoglobin and hemoglobin used as markers for screening of chicken meat quality in China

Abstract: A novel analytical protocol was developed for general quality screening of chicken meat based on IEF and protein extraction. To demonstrate the developed protocol, 24 chickens were divided into three groups; each had eight chickens. The chickens in Group 1 were slaughtered by exsanguination, Group 2 asphyxiated in water, and that in Group 3 were infected by new castle disease virus. Proteins were extracted from the meat samples by using pure water as an extractant, separated by IEF, verified by western blot, a… Show more

“…The samples were then analyzed using UPLC-MS/MS (Agilent model 1290). Mass spectrometric analyses were performed on an AB Sciex API 4000™ triple quadrupole mass spectrometer equipped with an electrospray ionization source to look for heterocyclic aromatic amines [15]. Separation was achieved on a Shim-pack GIST C18 (2.1 × 100 mm, 3 µm, 100 Å) at 37 • C. For further dilution, standard stock solutions containing 10 mg in 5 mL methanol were prepared.…”

“…However, the ruling principles of ion motion in solutions are totally different from those in a vacuum environment. , Due to strong viscous damping effects in solutions, ions usually no longer possess characteristic motional frequencies within a rf quadrupole electric field. Instead, the electrophoretic force (or dielectrophoretic force for particles) would be dominant in solutions. , Isotachophoresis (ITP), isoelectric focusing (IEF), and electric field gradient focusing (EFGF) methods have been developed and used primarily as separation techniques. In these methods, nonuniform solvents are required, so that either electric field gradients or electrolyte gradients could be generated inside liquid channels for ion separation.…”
