A Gradient of 2-Arachidonoylglycerol Regulates Mouse Epididymal Sperm Cell Start-Up1

Abstract: During transit through the epididymis, spermatozoa are normally kept immotile and do not attain the ability to become motile until they reach the caudal epididymis. This study was undertaken to determine whether endocannabinoids play a role in the epididymis and in particular in suppressing the ability of spermatozoa to become motile. We show that the levels of the endocannabinoid 2-arachidonoylglycerol (2-AG) are high in mouse spermatozoa isolated from the caput (head) of the epididymis, where these cells do … Show more

“…The involvement of the 'endocannabinoid system' in the development of sperm motility in the epididymis has recently been proposed (Cobellis et al 2010). The low levels of cAMP production induced by sAC from immature sperm may be related to the inhibitory effect on sAC exerted by the activated cannabinoid receptor CNR1 present on the sperm cell membrane.…”

“…In male, they act, directly and/or indirectly, at both central (hypothalamus and hypophysis) [16][17][18] and local levels (testis, epididymis and deferent ducts) via their cognate receptors [19][20][21][22][23][24][25][26]. At a central level, CNR1 regulates GnRH synthesis and release in frogs and rats, respectively [17,27].…”

“…An ECS has been demonstrated in human (Rossato et al 2005, Francavilla et al 2009), boar (Maccarrone et al 2005), mouse (Ricci et al 2007, Sun et al 2009), and bovine (Gervasi et al 2009). In mice, ECS modulates sperm motility during epididymal maturation, and high levels of endocannabinoids have been detected in mouse spermatozoa isolated from the caput of the epididymis, where they are functionally immotile, while low levels have been found in those isolated from the cauda where they acquire motility (Ricci et al 2007, Cobellis et al 2010. Inside the oviduct, the concentration of AEA is higher in the isthmus than in the ampulla.…”
