A large panel of novel microsatellite markers for the bank vole (Myodes glareolus)

Abstract: We describe a set of 66 highly polymorphic microsatellite loci isolated from the bank vole, Myodes (Clethrionomys) glareolus. These microsatellites were characterized for a long-term study on periodically fluctuating density of the bank vole population in Central Finland. We detected six to 38 alleles per locus in the population sampled at two different density phases, and the levels of observed and expected heterozygosities varied between 0.17 and 1.00, and between 0.72 and 0.95, respectively. This microsatel… Show more

“…Genomic DNA was extracted from tail tissue using the phenol-chloroform method. The following 17 microsatellite loci were amplified in three sets, using the Qiagen Multiplex Kit in a PTC-100TM (MJ Research) thermocycler: CG1E6, CG1E8, CG2A4, CG5F6, CG5G6, CG6D10, CG7C9, CG12A7, CG12B9, CG13F9, CG13G2, CG15F7, CG16E2, CG16E5, CG17E9 (Rikalainen et al 2008), MSCg-15 and MSCg-19 (Gockel et al 1997). The amplification conditions were 95°C for 15 min, followed by 30 cycles of denaturation at 94°C for 30 s, annealing at 57°C for 90 s and extension at 72°C for 60 s, and 60°C for 30 min.…”

“…To test whether TLR2 alleles behaved differently from neutral expectations, we genotyped the bank vole and yellow‐necked mouse samples at putatively neutral microsatellite loci. For bank voles, we used the following eight markers: Cg16E2, Cg13F9, Cg7E5, Cg3A8, Cg17A7, Cg12A7, Cg3A6, and Cg4F9 (Rikalainen et al 2008). Amplifications were performed in a 10 μl volume using Qiagen Multiplex Kit (Qiagen AB, Sollentuna, Sweden) with fluorescent‐labeled forward primers and nonlabeled reverse primers (0.2 μM each) on a GeneAmp PCR Systems 9700 thermocycler (Applied Biosystems).…”

“…We additionally genotyped bank voles at eight autosomal microsatellites, divided into two multiplexed sets (set 1: Cg16E2, Cg13F9, Cg7E5, Cg17A7, Cg3A8; set2: Cg4F9, Cg8A5, Cg1F11; Rikalainen et al, 2008;Tschirren et al, 2011) to estimate finescale genetic variation and genetic structure across locations. The amplification was carried out in a total volume of 10 lL, including 5 lL of 2 9 Qiagen Multiplex PCR Master Mix (Qiagen), 1 lL 10 9 primer mix and 20 ng of genomic DNA.…”
