A Method to Assess Histamine-Induced Cyclic AMP Production in Isolated Gastric Mucosal Cells from Human Biopsies

Abstract: Human gastric mucosal cells were isolated by digestion of fundic biopsies with pronase and collagenase. The mean value of gastric cells per milligram biopsy specimen ± SEM was 59,000 ± 4,300 (n = 31) with a viability of 90 ± 5 %. With the cell yield of 1 patient a series of approximately 70 – 80 cyclic AMP measurements was possible. Histamine stimulated intracellular cyclic AMP production with an EC50 value of 35 ± 25 μmol/l (SEM; n = 4). In the presence of 100 μmol/l histamine the Ki values (μmol/l) for the h… Show more

“…As a result, the combination of the choices of supportive bedding and enriched media, determine the resulting cellular composition (reviewed in Table 1), as compared to simple cell culture systems. 10,12,[14][15][16]18,46,[60][61][62] Therefore, we believe that our combination of MEF bedding and the 2D enriched media yield a cellular composition that includes epithelial, gastric-specific, and progenitor cells.…”

“…Gastric mucosal cells were prepared according to the methods of Mardh et al [30] and Sarem-Aslani et al [31] under sterile conditions. 5-7 biopsies were tak en endoscopically from antral mucosa and were trans ferred to a thermostated glass tube containing 3 mg pronase (from Streptomyces griseus: Boehringer, Mannheim) in 2 ml medium I (70 mmol/1 NaCl.…”
