Despite previous efforts, the evolutionary history of the immigrans-tripunctata clade remains obscure in part due to its hypothesized origin through a rapid radiation. We performed a supermatrix analysis (3,243 base pairs) coupled with richness patterns, environmental phylogenetic signal and radiation tests in order to address phylogenetic relationships and macro-evolutionary hypotheses within this complex group of species. We propose a well-supported evolutionary scenario for the immigrans-tripunctata clade species, in which the tripunctata "radiation" was monophyletic and subdivided into three main lineages: the first including D. pallidipennis (pallidipennis group) imbedded among members of the tripunctata group; the second clustering the cardini and guarani groups; and the third grouping representatives from the tripunctata, calloptera and guaramunu groups. Therefore, we hypothesize that the tripunctata group encompasses a diphyletic taxon, with one clade including the pallidipennis group and the other showing a close affinity to the calloptera and guaramunu groups. Our results also suggest that niche evolution seems to have played a central role in the evolutionary history of the tripunctata species "radiation" allowing effective dispersion and diversification in the Neotropics, possibly in a southwards direction. Although the data as a whole support the notion that this occurred through rapid and successive speciation events, the radiation hypothesis remains to be further corroborated.

The location of rDNA on chromosomes has been found to be highly variable in different groups of insect families of various orders. Yet, in other insect families the rDNA position is relatively constant. This contrast so far has received limited attention. We investigated the rDNA position on mitotic chromosomes in 18 species of Drosophila, 16 of which are from the tripunctata radiation, subgenus Drosophila, through fluorescent in situ hybridization (FISH). All species showed fluorescent signals only on the sex chromosomes. On the X chromosome a single fluorescent mark, but with variable locations, was found. On the Y, we observed variation both in location and in number of fluorescent marks (from 1 to 5). This constancy of chromosome location, in contrast to the great variability found in other groups, is consistent with the work carried out in other species of Drosophila. This suggests that the presence of a nucleolus organizer region (NOR) on each sex chromosome is probably an ancestral condition in the genus. Moreover, this difference in the variation of rDNA position among groups points out an interesting evolution question, which deserves further study.

Se realizó un muestreo a lo largo de la Quebrada de Cruz Loma, un remanente de Bosque Andino ecuatoriano, situada entre los 3100 y los 4000 m de altitud. Se capturaron 718 individuos del género Drosophila, entre ellos a una nueva especie miembro del grupo de especies de Drosophila tripunctata: Drosophila condorhuachana sp. nov. Con esta nueva especie el número de especies del grupo de D. tripunctata registradas en el Ecuador asciende a 22.
