A multifunctional nanoplatform for cancer chemo-photothermal synergistic therapy and overcoming multidrug resistance

Abstract: The integration of various therapy strategies into a single nanoplatform for synergistic cancer treatment has presented a great prospect. Herein, docetaxel (DTX)-loaded poly lactic-co-glycolic acid (PLGA)-coated polydopamine modified with d-α-tocopherol polyethylene glycol 1000 succinate (TPGS) was synthesized for chemo-photothermal synergistic therapy against cancer. Firstly, the DTX-loaded PLGA NPs were prepared by a facile and robust nanoprecipitation method. Then, they were coated with dopamine to achieve … Show more

“…and polymeric NPs with NIR-absorbing agents like ICG ( Cheng et al, 2014 ). In that line PLGA and its copolymers (e.g., PLGA-PEG, PLGA-lipid) have been used for co-loading and combination therapy with, e.g., DTX and gold (Au) ( Hao et al, 2015 ), DTX and poly(dopamine) ( Peng et al, 2018 ) or DTX and polypyrrole (PPy) ( Yuan et al, 2016 ). The latter PTT agent, PPy, is considered an ideal heat inductive material, which was used by Yuan et al to build a DOX encapsulated PLGA-PPy NP.…”

“…It is well known that NPs coated with PEG can accumulate in different types of solid tumors due to the EPR effect; they are considered suitable vehicles for hydrophobic drugs, able to attain efficient tumor targeting with the fewest adverse reactions (Venkatasubbu et al., 2013 ; Siqi et al., 2019 ). Several methods were applied for NPs development including nanoprecipitation method (Peng et al., 2018 ), solvent evaporation method (Catarina et al., 2006 ), dialysis (Rao & Geckeler, 2011 ), and salting out (Sovan et al., 2011 ). Brucine (BRU) is a white, odorless, crystalline, and poorly water-soluble anticancer drug extracted from Strychnos nux-vomica seeds (Gupta & Chaphalkar, 2015 ).…”

“…PCL is taken as a polymeric membrane as a drug carrier since it has been extensively used in biomedical purposes owing to its excellent biocompatibility, tunable mechanical properties and slow biodegradation 3134 . Although PDA has been widely applied in particulate drug delivery systems and hydrogels for photothermal therapy 1419,24,3538 , to the best of our knowledge the potential functional use of PDA in localized drug delivery using nanofibrous membranes has not yet been reported. The present study aims to develop smart multiple stimuli-responsive nanofibers that are capable of both pH/NIR dependent drug release in a cancer environment and heat generation under the application of NIR triggered phototherapy for the combined treatments of hyperthermia and chemotherapy.…”
