A needle in a seedstack: an improved method for detection of rare alleles in bulk seed testing through KASP

Abstract: BACKGROUND Amaranthus palmeri is an aggressive and prolific weed species with major impact on agricultural yield and is a prohibited noxious weed across the Midwest. Morphological identification of A. palmeri from other Amaranthus species is extremely difficult in seeds, which has led to genetic testing for seed identification in commercial seed lots. “…Monitoring for movement of species and for occurrence of resistance within species is a key component of integrated weed management, requiring accessible and accurate diagnostic methods. Genotypic diagnostics for species identification can improve monitoring of weeds such as A. palmeri in the trade of seeds and grain, such as the recently reported Kompetitive Allele Specific PCR (KASP) assay 23 and qPCR assay 24 for A. palmeri identification, 23 thereby helping to monitor for and prevent new introductions. Our ability to rapidly diagnose herbicide resistance is improving, including for NTSR.…”
