A Neutralizing Antibody Selected from Plasma Cells That Binds to Group 1 and Group 2 Influenza A Hemagglutinins

Abstract: The isolation of broadly neutralizing antibodies against influenza A viruses has been a long-sought goal for therapeutic approaches and vaccine design. Using a single-cell culture method for screening large numbers of human plasma cells, we isolated a neutralizing monoclonal antibody that recognized the hemagglutinin (HA) glycoprotein of all 16 subtypes and neutralized both group 1 and group 2 influenza A viruses. Passive transfer of this antibody conferred protection to mice and ferrets. Complexes with HAs fr… Show more

“…There is a growing interest in passive transfer of anti-influenza Abs in severe influenza, with some efficacy observed in small human studies (43,44) and animal models (45). Both polyclonal immunoglobulin preparations (28) and some monoclonal Abs can have significant Fc-mediated functions across a range of influenza strains (24,25,33). Future interventional studies complementing anti-influenza ADCC in subjects with severe influenza are warranted.…”

“…This is especially promising with similar approaches in HIV, 21,22 HCV 23 and influenza. 24 The prophylactic administration of bovine lactoferrin or probiotic fermented milk failed at preventing NoV infection in children and elderly residents of a health care facility, respectively. 25,26 The development of an effective treatment for NoV infections among children and immunosuppressed patients is needed to reduce the burden of gastroenteritis.…”

“…Progress is being made in this area to avoid having to constantly update vaccines. Recently, researchers have identified broadly neutralizing human antibodies against influenza haemagglutinin, which could be exploited to develop a universal flu vaccine (Corti et al, 2011;Ekiert et al, 2009Ekiert et al, , 2011. However, for organisms that exhibit very substantial genetic diversity and variation, particularly in (Moxon & Siegrist, 2011).…”
