A new iridoid glycoside and NO production inhibitory activity of compounds isolated from Russelia equisetiformis

Abstract: From the 1-BuOH-soluble fraction of a MeOH extract of the leaves of Russelia equisetiformis, one new iridoid glucoside was isolated along with 24 known compounds, comprising iridoids and iridoid glucosides, phenyl propane glucosides, phenyl ethanoids, lignan glucosides, and flavonoid glucosides. The structure of the new compound was elucidated to be 10-O-cinnamoyl sinuatol. Of the 25 compounds isolated, rehmaglutin B exhibited moderate inhibitory activity toward NO production, which was not associated with cyt… Show more

“…It is reported that the plant is traditionally used in Nigeria for the treatment of diabetes, leukemia and pain and inflammation. Scientific studies have revealed analgesic, anti-inflammatory, antimicrobial, free radical scavenging and membrane-stabilising activity [134-136]. Whole organic plant extracts and isolated phenolic compounds exhibited antinociceptive activity in male Swiss mice [134] in both the writhing and tail-flick tests.…”

“…The isolated compounds, including rehmaglutin B ( 37 ), catalpol ( 15 ), 6- O - cis -p-coumaroylcatalpol, and verminoside ( 25 ) amongst others were assayed for their inhibitory activity towards NO production in vitro using RAW 264.7 cells. The iridoid compounds verminoside (±8%) ( 25 ) and rehmaglutin B (70%) ( 37 ) showed NO inhibitory activity at 100 µM and rehmaglutin B (IC 50 = 52.8±2.0 µM) inhibited NO production more strongly than the control (L-NMMA) [136]. …”

“…Vien et al [ 14 ] obtained a different result in LPS-stimulated BV2 microglial cells using the Griess assay, with an IC 50 value of 61.1 µM, which is much higher than the positive control butein (IC 50 = 4.5 µM). In contrast, Ochi et al [ 147 ] obtained a moderate percentage of inhibition of 40% in cell culture supernatants of LPS-stimulated RAW264.7 macrophages at the concentration of 100 µM, whereas the positive control L-NMMA presents an inhibition value of 55% at the same concentration.…”

“…The positive control butetin had much stronger effects (IC50 = 4.5 µM). Lastly, Ochi et al [145] also obtained moderate results in cell culture supernatants of LPS-stimulated RAW264.7 macrophages with a percentage of inhibition of 40%.…”
