Leptin (LEP) gene produces hormone leptin which is secreted by adipose tissue, and plays an im-portant role in energy balance, regulating feed intake, regulating endocrine function, and immune functions. This study aims to identify molecular markers of LEP gene and its association with growth traits based on SNP in Bali cattle. The blood samples were collected from 16 male and 30 female Bali cattle. The growth data were recorded from 2018 to 2020, consist of body weight and body size. Am-plification of leptin gene with polymerase chain reaction (PCR) using pair of primers, Lep-tin_3Forward: 5’- AGCTTGGAAACATGGTGGTC-3’ and Leptin_3Reverse: 5’- CATGATGCTCCCTGGATTCT-3’ with DNA target 898 bp. The SNPs were identified by the direct sequencing technique. Genotypes of the SNPs were identified using sequencing method. Association of LEP genotypes with growth traits was performed using oneway ANOVA. Three DNA polymor-phisms of the LEP gene were found, namely g.2913C/T, g.3260T/C, and g.3549G/A. SNP g.2913C/T was significantly associated (P<0,05) with weaning shoulder height (WSH), weaning body length (WBL), weaning chest circumference (WCC), yearling shoulder height (YSH), yearling body length (YBL), and yearling chest circumference (YCC). Meanwhile, SNPs g.3260T/C and g.3549G/A were not associated with the body weight and body size in Bali cattle. In conclusion, the SNP g.2913C/T can be used as molecular marker for body size in weaning and yearling of Bali Cattle.

This study aimed to investigate the genetic impact of single nucleotide polymorphisms (SNPs) of the sterol regulating element binding factor 1 (SREBF1) and scavenger receptor class B member 1 (SCARB1) genes on carcass and meat characteristics, as well as fatty acid composition, in the Bali cattle. The blood and beef samples used for DNA sequencing, physical assessment, and fatty acid analysis were collected from 95 male Bali cattle. The ultrasound images were analyzed using the Image-J NIH software. A total of 4 SNPs were identified in the SREBF1 gene and 5 SNPs in the SCARB1 gene. The results showed that the 4 SNPs in the SREBF1 gene, namely g.12629T>C, g.12731T>C, g.12881A>G, and g.12986C>T, were associated with heptadecanoic acid (C17:0) and cis-11-eicosanoic acid (C20:1). The SNPs g.12731T>C of the SREBF1 gene was associated with fat content, palmitoleic acid (C16:1), stearic acid (C18:0), cis-11-eicosanoic acid (C20:1), and total fatty acids. Furthermore, 4 SNPs in the SCARB1 gene, including g.72219C>T, g.72380C>A, g.72517G>A, and g.72607C>T correlated with longissimus dorsi thickness (LDT). All SNPs in the SCARB1 gene showed significant associations with cis-10 heptadecanoic acid (C17:1) and cis 8,11,14-eicosatrienoic acid (C20:3n6). The SNP g.72400A>G of the SCARB1 gene was related to caprylic acid (C8:0), lauric acid (C12:0), arachidonic acid (C20:4n6), monounsaturated fatty acids (MUFA), and unsaturated fatty acids (UFA). These results suggested that the identified polymorphisms in the SREBF1 and SCARB1 genes could serve as valuable references for investigating similar genes in other cattle breeds, particularly concerning fatty acids.

Abstract. Calpain 1 (CAPN1) is an enzyme that influences meat tenderization, and it is involved in post mortem proteolysis. The bovine CAPN1 gene was chosen as a candidate gene for DNA sequencing to identify novel single-nucleotide polymorphisms (SNPs) in exons 8–10 and assess their associations with meat quality in Bali cattle. In an analysis of 95 Bali cattle, two novel SNPs (g.5327C>T and g.5959C>T) were identified in exons and four novel SNPs (g.5534C>T, g.5807A>C, g.5857G>A, and g.5869T>C) were discovered in introns. Allelic frequency was evaluated, and Hardy–Weinberg equilibrium was found for all SNPs in Bali cattle. The marbling score and intramuscular fat content as determined by ultrasound were associated with g.5869T>C and g.5959C>T. g.5327C>T was associated with pH and meat color in Bali cattle, whereas two other SNPs (g.5869T>C and g.5959C>T) were associated with Warner–Bratzler shear force and meat color. Furthermore, g.5869T>C was associated with linolenic acid content, and g.5959C>T with caprylic and linolenic acid levels. However, intronic SNPs (g.5534C>T, g.5807A>C, and g.5857G>A) did not significantly affect meat quality in Bali cattle. Quantitative real-time PCR of liver tissue revealed that the mRNA expression of CAPN1 significantly differed (P<0.05) among the CT, CC, and TT genotypes. The results suggest that genetic variability in loci within CAPN1 might be associated with meat quality in Bali cattle.
