A novel recombinant vaccine protecting mice against abrin intoxication

Abstract: Abrin toxin (AT) consisting of an A chain and a B chain is a potential agent for bioterrorism and an effective vaccine against AT poisoning is urgently required. In this study, AT B chain (ATB) was successfully expressed in the Escherichia coli (E. coli) and assessed the protection capacity against AT intoxication. The recombinant ATB (rATB) subunit induces a good immune response after 4 immunizations. All BALB/c mice immunized intraperitoneally (i.p.) with the purified rATB protein survived after challenged w… Show more

“…Abrin is a highly toxic protein with a relative molecular mass of 65 kDa 34 that is composed of an A-chain and a B-chain. The estimated lethal oral dose of abrin in humans is 0.1–1 μg/kg 35 , 36 . Unintentional abrin ingestion can quickly cause irreparable organ failure and even death, so rapid, label-free, and sensitive detection of abrin is desired.…”

“…Future protection efforts are aimed at developing a vaccine against inhaled ricin and abrin. No approved vaccine or specific antidote is currently available for human use, but experimental, recombinant vaccines are under development (Pittman et al 2015;Wang et al 2015). If exposure to skin occurs, decontamination with soap and water is available.…”

“…In mice, generation of protective antibodies can be induced with a sub-lethal dose of abrin (10,11). Recombinant abrin A chain (12)(13)(14)(15) or the weaker abrin B chain (16)(17)(18)(19), can also induce anti-abrin antibody production. Up to now, only two neutralizing antibodies against Abrin poisoning have been reported, D6F10 and A7C4, but neither has entered clinical trials or been used directly in humans.…”
