A Peroxisome‐Inspired Chemiluminescent Silica Nanodevice for the Intracellular Detection of Biomarkers and Its Application to Insulin‐Sensitizer Screening

Abstract: The profiling of oxidase-catalyzed biomarkers is an essential procedure for the diagnosis and precise treatment of metabolic diseases. Inspired by the metabolism of H O in peroxisomes, a novel chemiluminescent silica nanodevice (CSN) was designed for the sensitive and selective sensing of intracellular oxidase-catalyzed biomarkers. Oxidases catalyzed the oxidation of biomarkers followed by the production of H O , and then the generated H O was employed to trigger chemiluminescence of the CSN. Utilizing this na… Show more

“…[11] It is one of the most important characteristics related to cell spreading area, cell size,c ell activity,a nd intercellular metabolites. [12] Current methods for cell adhesion measurement include protein staining, [13] fluorescein labeling, [14] isotope-labelling, [15] and intracellular enzyme release assays [16] that are carried out by testing the adhered cells on asurface. Thus, the cell adherence ability was characterized by counting the number of cells, which yields statistical results that cannot represent the cell adherence ability of ap articular single cell. Thus,t he measurement of single-cell adherence ability and connecting it to cell viability is still av ital issue.…”

“…Although there are several approaches for cell adhesion measurement, [12][13][14][15] none of them allowed the measurement at single-cell resolution and the distinction of the individual differences between different cells.T he U87 cells at different positions in the petri dish were extracted one by one (Figure 3d), and the time required for extraction was measured at the same time.T he area, Feretsd iameter,a nd perimeter of each single cell were analyzed using Image-Pro plus software (Media Cybernetics Inc.,Bethesda, MD,USA). TheP earson correlation coefficient (PCC) was calculated using IBM SPSS Statistics software 22.0 (SPSS Inc.,Chicago, IL, USA).…”

“…The electrostatically adsorbed oxidase catalyses the production of peroxide and further protects the peroxyoxalate from degradation. Upon cell uptake, biomarkers such as glucose, lactic acid, uric acid and ethanol, which are catalytically reacted to H 2 O 2 , were quantified by chemiluminescence emission in living mice (refer to Scheme ) …”
