A Phytophthora sojae effector suppresses endoplasmic reticulum stress-mediated immunity by stabilizing plant Binding immunoglobulin Proteins

Abstract: Phytophthora pathogens secrete an array of specific effector proteins to manipulate host innate immunity to promote pathogen colonization. However, little is known about the host targets of effectors and the specific mechanisms by which effectors increase susceptibility. Here we report that the soybean pathogen Phytophthora sojae uses an essential effector PsAvh262 to stabilize endoplasmic reticulum (ER)-luminal binding immunoglobulin proteins (BiPs), which act as negative regulators of plant resistance to Phy… Show more

“…'Protein Processing' is represented by proteins with molecular functions in 'Ubiquitination', 'Protein Folding/Stability', 'Protein Degradation' and 'Protein Sorting'. For example, the Phytophthora sojae effector PsAvh262 targets multiple soybean BiP family of chaperones and the Magnaporthe oryzae effector AvrPiz-t targets multiple E3 ligases (APIPs; Park et al, 2012;Jing et al, 2016;Park et al, 2016;Tang et al, 2017). In terms of molecular functions, while bacteria predominantly target 'Kinases' (32%), oomycete targets are more diverse in molecular function, including 'Oxidoreductases' (19%), 'Protein Folding/Stability' (17%) 'Phosphatases' (14%), or 'Promoters' (14%), with 'Kinases' only representing 2% of the total (Figure 4(d)).…”

“…For P. sojae, RNA-seq data were obtained at different stages of the life cycle including the mycelium, zoospore, and germinated cyst, as well as soybean roots (cultivar: Williams 82) at 3, 6, 12, 24 and 48 h after inoculation with P6497 zoospores, as previously described (Jing et al, 2016). Similarly, P. capsici RNA-seq data were obtained at various stages of the life cycle including the mycelium, sporangium, zoospore, and germinated cyst, as well as pepper roots (cultivar: EC01) at the indicated time points after inoculation with JX1 zoospores (W. Cheng, unpublished).…”

“…Interfering with the stability of target proteins or crucial immune components is commonly used by pathogen effectors. For instance, the P. sojae RxLR effector PsAvh262 targets and stabilizes BiPs (binding immunoglobulin proteins) to suppress ER stress‐mediated immunity and facilitate infection (Jing et al , ). The Xanthomonas oryzae pv.…”
