A prognostic classification system for uveal melanoma based on a combination of patient age and sex, the American Joint Committee on Cancer and the Cancer Genome Atlas models

Abstract: Uveal melanomas (UMs) most commonly arise in the choroid (90% of cases), followed by the ciliary body (6%) and iris (4%) (Shields et al., 2009). Whereas iris melanomas are distinguished by their relatively favourable prognosis and genomic features associated with ultraviolet radiation damage, choroidal and ciliary body melanomas have a higher propensity for metastatic spread and are likely initiated by punctuated evolution oncogenic events

“…This study also highlighted the statistical significance of ciliary body involvement (CBI) in prognosis. Concerning therapeutic implications, Gill VT. et al's study reported the absence of metastasis for the first favorable prognostic class, while Group 4 patients had a very high metastatic risk, and this may warrant inclusion in randomized trials with adjuvant treatment [49].…”

“…Gill VT. et al's 2023 study proposed the incorporation of an improved classification system based on patient age, gender, AJCC T-categories, and TCGA groups A to D [49]. This study also highlighted the statistical significance of ciliary body involvement (CBI) in prognosis.…”

“…The TCGA project was established with the primary objective of comprehensively characterizing the molecular alterations inherent in cancer cells through the analysis of vast amounts of data acquired from numerous human samples. This ambitious initiative aimed to enhance our understanding of the intricate molecular landscape of cancer and provide valuable insights into the underlying mechanisms driving tumorigenesis and disease progression) [8,9,14,122] Abbreviations: BAP1 BRCA1 Associated Protein 1, SF3B1 Splicing Factor 3b Subunit 1, EIF1AX Eukaryotic Translation Initiation Factor 1A X-Linked to induce tolerance in homeostasis [126,127].
