“…In contrast, COX-2 knock-out mice are more resistant to tumorigenesis in various organs (Oshima et al, 1996;Tiano et al, 2002;Howe et al, 2005). A number of clinical studies were performed for colorectal cancer prevention by administration of Nonsteroidal anti-inflammatory drugs (NSAIDs) or COX-2 inhibitors including sulindac, aspirin, celecoxib, rofecoxib (Giardiello et al, 1996;Steinbach et al, 2000;Giardiello et al, 2002;Baron et al, 2003;Higuchi et al, 2003;Sandler et al, 2003;Bertagnolli et al, 2006). These COX-2 inhibitors reduced adenoma only at a high dose but significantly increased mortality from cardiovascular complication.…”

“…Numerous studies demonstrate that nonsteroidal anti-inflammatory drugs (NSAIDs) hold significant promise as anticancer therapeutics (10)(11)(12). Recent data on regular NSAID use indicated a reduction in the incidence of OC and inhibition of carcinogenesis (12)(13)(14)(15).…”

“…Undoubtedly, several recent clinical results validate the conceptual feasibility of cancer chemoprevention in humans. These findings bear out that nonsteroidal anti-inflammatory drugs such as aspirin (Baron et al, 2003) or selective inhibitors of cyclooxygenase-2 (COX-2) (Steinbach et al, 2000) interfere with colorectal malignancies, that selective oestrogen receptor modulators such as tamoxifen can prevent breast cancer (Fisher et al, 1998) and that the a-reductase inhibitor finasteride can delay the onset of prostate cancer (Thompson et al, 2003). However, chemopreventive interventions using drugs have raised safety concerns.…”
