“…Figure 1 shows the IR spectra of PHB before and after processing by extrusion. The characteristic absorption bands of PHB can be seen at 2928 cm -1 corresponding to the methylene C-H vibration, the symmetric -C-O-C-stretching vibration in the range of 800-975 cm -1 and the C=O stretching at 1722 cm -1 [16] . There are many studies regarding the use of FT-IR technique to determine the crystallization degree of PHB by monitoring the C=O stretching vibrational region (1780-1700 cm -1 ) [17,18] .…”

“…Hong et al developed a rapid noninvasive technique using Fourier Transformed Infrared Spectroscopy (FT-IR) which allows detecting intracellular PHA accumulation in intact cells within a few seconds. If the PHA content is high in the cells, the FTIR spectra of PHA can also differentiate the various structures of PHB (scl), mcl-PHAs and PHA consisting of HB and mcl-3HA monomers (80). Genotypic-based screening method is specific, reliable and quick which circumvents the major drawbacks inherent in phenotypic detection methods (78,(81)(82)(83)(84).…”

“…For genotypic-based screening, the degenerate or and specific oligo nucleotide primers are designed based on multiple sequence alignment results and are used as PCR primers to detect PHA synthase genes (78,79,81,(83)(84)(85). For rapid detection, colony PCR technique is employed for screening PHA producers from the environment (78,80). The PHA accumulation ability of well-separated colonies isolated from environmental samples can be directly validated by PCR with no further culturing or chromosomal DNA extraction procedures.…”

“…PHA accumulation after 24h was 23 wt% of cell dry mass while after 72 h of incubation it reached to 42.3 wt% of cell dry mass. Purified PHA showed strongest band at 1735 cm -1 , in comparison, to PHA being produced A. zotobacter vinelandii and Pseudomonad mendocina their carbonyl bands (C=O) at 1732 cm -1 (Hong et al, 1999). In the presence of sodium gluconate mcl-PHA was identified by H + NMR, hence it was concluded that CMG607w accumulated a polymer with medium chain length as the major constituent when the cells were grown on sodium gluconate (Huijbert et al, 1992).…”
