Diosgenin Derivatives as Potential Antitumor Agents: Synthesis, Cytotoxicity, and Mechanism of Action

Thirty-two new diosgenin derivatives were designed, synthesized, and evaluated for their cytotoxic activities in three human cancer cell lines (A549, MCF-7, and HepG2) and normal human liver cells (L02) using an MTT assay in vitro. Most compounds, especially 8, 18, 26, and 30, were more potent when compared with diosgenin. The structure−activity relationship results suggested that the presence of a succinic acid or glutaric acid linker, a piperazinyl amide terminus, and lipophilic cations are all beneficial for promoting cytotoxic activity. Notably, compound 8 displayed excellent cytotoxic activity against HepG2 cells (IC 50 = 1.9 μM) and showed relatively low toxicity against L02 cells (IC 50 = 18.6 μM), showing some selectivity between normal and tumor cells. Studies on its cellular mechanism of action showed that compound 8 induces G0/G1 cell cycle arrest and apoptosis in HepG2 cells. Predictive studies indicated that p38α mitogen-activated protein kinase (MAPK) is the optimum target of 8 based on its 3D molecular similarity, and docking studies showed that compound 8 fits well into the active site of p38α-MAPK and forms relatively strong interactions with the surrounding amino acid residues. Accordingly, compound 8 may be used as a promising lead compound for the development of new antitumor agents.

There are several Amazonian plant species with potential pharmacological validation for the treatment of acute kidney injury, a condition in which the kidneys are unable to adequately filter the blood, resulting in the accumulation of toxins and waste in the body. Scientific production on plant compounds capable of preventing or attenuating acute kidney injury—caused by several factors, including ischemia, toxins, and inflammation—has shown promising results in animal models of acute kidney injury and some preliminary studies in humans. Despite the popular use of Amazonian plant species for kidney disorders, further pharmacological studies are needed to identify active compounds and subsequently conduct more complex preclinical trials. This article is a brief review of phytocompounds with potential nephroprotective effects against acute kidney injury (AKI). The classes of Amazonian plant compounds with significant biological activity most evident in the consulted literature were alkaloids, flavonoids, tannins, steroids, and terpenoids. An expressive phytochemical and pharmacological relevance of the studied species was identified, although with insufficiently explored potential, mainly in the face of AKI, a clinical condition with high morbidity and mortality.

Obesity and cholesterol are very feared problems. In recent years, many people have tried to use alternative treatments by consuming ingredients from plants, one of which is telang flower tea (Clitoria ternatea). The purpose of this study was to test and determine the difference in the effect of the frequency of tea drinks (Clitoria ternatea) accompanied by aerobic exercise on weight loss (Body Mass Index) and cholesterol. Experimental research design with pre and post-test design methods. Group grouping using matching pairing and data collection took measurements of body mass index, and blood tests in the form of cholesterol tests. The study population was all Military National Indonesian members who had a BMI > 25. The data analysis technique uses a difference test of 2 paired groups to distinguish the pretest and posttest in each group, with a significance level of 5%. The results of the analysis showed that: 1) there was an effect of the frequency of drinking 2 times the tea accompanied by aerobic exercise on the decrease in Body Mass Index (BMI) and cholesterol, 2) There was an effect of the frequency of drinking 5 times the tea accompanied by aerobic exercise on the decrease in Body Mass Index (BMI) and cholesterol, 3) There was a difference in the effect of the frequency of drinking 2 times and 5 times the tea accompanied by aerobic exercise on reducing Body Mass Index (BMI) and cholesterol.
