Background and Objective: Fruits are important sources of income for farmers. However, they are subject to many diseases caused mainly by fungi, of which the genus Colletotrichum. The genus Colletotrichum is an important and frequent causal agent of post-harvest infections of fruits. Although this genus can infect a wide range of hosts, one or more species of Colletotrichum may be found on the same host. Based on morphological characteristics, this study aimed to differentiate 49 isolates of Colletotrichum sp., obtained from avocados and mangoes from the main production areas in Côte d'Ivoire. Materials and Methods: In this study, the growth rate of Colletotrichum isolates was evaluated on three culture media (potato dextrose agar (PDA), malt agar and corn meal agar (CMA) media), as well as their sporulation. The size of the spores was measured and their shapes were noted. Descending hierarchical classification (DHC) was used to group the isolates by size. A comparison of isolates obtained from the two fruits was performed by Principal Component Analysis (PCA) based on all morphological characteristics studied. Results: The study revealed a very high morphological variability in the conidia of Colletotrichum isolates. DHC grouped avocado isolates into 11 classes and mango isolates into four classes. Spore shape analysis revealed a majority of spores with either both ends rounded or one end rounded and the other sharp. Growth and sporulation of the isolates were best on the malt medium. PCA revealed that mango isolates were distinct from avocado isolates. Conclusion: This study showed that most isolates had cylindrical or ellipsoidal spores with rounded or one sharp end, with better growth and sporulation on the malt medium. However, these isolates were distinct and reported to have characteristics of Colletotrichum gloeosporioides and Colletotrichum acutatum.

Although mechanisms involved in response of Saccharomyces cerevisiae to osmotic challenge are well described for low and sudden stresses, little is known about how cells respond to a gradual increase of the osmotic pressure (reduced water activity; a w) over several generations as it could encounter during drying in nature or in food processes. Using glycerol as a stressor, we propagated S. cerevisiae through a ramp of the osmotic pressure (up to high molar concentrations to achieve testing-to-destruction) at the rate of 1.5 MPa day-1 from 1.38 to 58.5 MPa (0.990-0.635 a w). Cultivability (measured at 1.38 MPa and at the harvest osmotic pressure) and glucose consumption compared with the corresponding sudden stress showed that yeasts were able to grow until about 10.5 MPa (0.926 a w) and to survive until about 58.5 MPa, whereas glucose consumption occurred until 13.5 MPa (about 0.915 a w). Nevertheless, the ramp conferred an advantage since yeasts harvested at 10.5 and 34.5 MPa (0.778 a w) showed a greater cultivability than glycerol-shocked cells after a subsequent shock at 200 MPa (0.234 a w) for 2 days. FTIR analysis revealed structural changes in wall and proteins in the range 1.38-10.5 MPa, which would be likely to be involved in the resistance at extreme osmotic pressure.

Huge consumption of fossil fuel energy creates environmental problems, while the amount of organic solid waste is in an increasing trend especially in developing countries. Biogas is known as a renewable energy that can be used as an alternative fuel which is promising to reduce dependency on fossil fuels. This study identified the concentration of methane gas that can be produced from organic waste treated in an anaerobic digestion with the addition of Hyphomicrobium sp. Two-stage batch anaerobic digestion was used with an operational volume of 6 L. Four different starting concentrations of bacteria Hyphomicrobium sp. added to the reactors: 0.29 × 10 9 cells/L in reactor A, 0.87 × 10 9 cells/L in reactor B, 1.75 × 10 9 cells/L in reactor C, and no Hyphomicrobium sp. in the control reactor. The results showed that the concentration of methane gas produced was 38.4% in the reactor A while in reactor B and C were 35.6% and 33%, respectively. In reactor K with no addition of bacteria, the amount of methane produced was 52%. In contrary to the hypothesis, the addition of Hyphomicrobium sp. to the anaerobic process was contra-productive to the yield of methane. Further research is required to investigate the role of Hyphomicrobium sp. on the methane gas formation in the reactor.
