A sensitive LC‐MS/MS method for the simultaneous determination of amoxicillin and ambroxol in human plasma with segmental monitoring

Abstract: Amoxicillin (AMO) degrades in plasma at room temperature and readily undergoes hydrolysis by the plasma amidase. In this paper, a novel, rapid and sensitive LC-MS/MS method operated in segmental and multiple reaction monitoring has been developed for the simultaneous determination of amoxicillin and ambroxol in human plasma. The degradation of amoxicillin in plasma was well prevented by immediate addition of 20 μL glacial acetic acid to 200 μL aliquot of freshly collected plasma samples before storage at -80°C… Show more

“…Generally, drug analysis can't do without high performance liquid chromatography or capillary electrophoresis or LC-MS/MS or NMR et al The metabolic profiling and metabolic fingerprinting applications can be addressed as well as pharmacodynamic and pharmacokinetic issues from model organisms, animals and humans [10]. HPLC [11][12][13], LC-MS and LC-MS/MS [14] methods were applied to the drug analysis in various biological matrices for the pharmacokinetic mechanism. These methods are time-consuming and labor-intensive for the therapeutic application [15,16].…”

“…The AMX extraction method was modified from those previously described (Wen et al ., ; Dong et al ., ). Briefly, 200 μL of plasma sample was added with 20 μL of glacial acetic acid in a 2‐mL plastic centrifuge tube.…”

“…5 Some bioanalytical methods have been reported for the determination of ambroxol by HPLC-MS/MS analysis. [6][7][8][9][10][11] In these methods, electrospray ionization (ESI) was most commonly used as an ionization method. However, in our preliminary experiments, ESI had high matrix effects for determining ambroxol in human plasma.…”
