A serine proteinase inhibitor isolated from Tamarindus indica seeds and its effects on the release of human neutrophil elastase

Abstract: Proteinaceous inhibitors with high inhibitory activities against human neutrophil elastase (HNE) were found in seeds of the Tamarind tree (Tamarindus indica). A serine proteinase inhibitor denoted PG50 was purified using ammonium sulphate and acetone precipitation followed by Sephacryl S-300 and Sephadex G-50 gel filtration chromatographies. Inhibitor PG50 showed a Mr of 14.9 K on Sephadex G-50 calibrated column and a Mr of 11.6 kDa on sodium dodecyl sulfate-polyacrylamide gel electrophoresis. PG50 had selecti… Show more

“…Till date trypsin inhibitors have been purified and characterized from a variety of plant sources (Macedo et al2000;Oliviera et al 2007;Kansal et al 2008;Rai et al 2008). The purified inhibitor protein was found to consist of a single polypeptide chain of 12,303 daltons as revealed by Native-PAGE and SDS-PAGE, which is consistent with the molecular masses of other trypsin inhibitors (Mizuta and Ventura 1976;Fook et al 2005;Sivakumar et al 2005). On the contrary trypsin inhibitor purified from A. kalkora seeds showed the presence of two disulfide linked polypeptide chains and a high molecular mass (Zhou et al 2008).…”

“…It is clear that the inhibitory actions of the hawthorn extracts on prolyl endopeptidase are not attributable to these low molecular weight phenolic compounds. For another common protease, elastase, which is distributed in various tissues, such as the pancreas, lung, and neutrophils, 42) the results shown in Table 2 indicate that hawthorn extracts do not have significant inhibitory action. This indicates that the inhibitory action of the hawthorn extracts on prolyl endopeptidase is selective.…”

“…Water extract of tamarind seed has been shown to reduce blood sugar level in Streptozotocin-induced diabetic male rats (Maiti et al 2004). Tamarind seeds are known to have high inhibitory activity against human neutrophil elastase (Fook et al 2005).…”
