A Simple Extraction and Chromatographic System for the Simultaneous Analysis of Anthocyanins and Stilbenes ofVitisSpecies

Abstract: A separation system has been developed to permit the simultaneous analysis of major anthocyanins and stilbenes from berries of Vitis species in <1 h. The system makes use of separation by HPLC and detection by UV-visible absorption and fluorescence, for anthocyanins and stilbenes, respectively, with the two detection systems linked in series. Monitoring the absorption at 520 nm permits ready identification and quantification of major anthocyanins. Chromatograms derived from fluorescence (330 nm excitation/374 … Show more

“…, 2002), research efforts to characterize its regulation and the location of its biosynthesis and accumulation has increased. It is generally accepted that trans ‐ and cis ‐resveratrol and their respective 3‐ O ‐monoglucosides ( trans ‐ and cis ‐piceid) are the major stilbenes that accumulate in grape exocarp tissue (Ali and Strommer, 2003; Versari et al. , 2001).…”

“…, 2001). Vitis vinifera accumulates the majority of its stilbenes in the berry exocarp (1.69–8.69 μ g per berry skin), with the amount and type of stilbene (glucosylated or not) that accumulates being cultivar‐specific (Ali and Strommer, 2003). In addition, it has been shown that UV irradiation of Vitis vinifera (cv.…”

“…A slow gradient at the beginning of the chromatogram was necessary to allow a total separation of the transresveratrol peak from other polyphenols. In our micro-LC system trans-resveratrol eluted with a lower percentage of organic solvent with respect to other classical columns [19][20][21][22]30], but the efficiency was good enough to completely isolate its peak.…”

“…Grape berries accumulate the 3-O-monoglucosides and the 3,5-O-diglucosides of the colorful anthocyanins (Muñoz-Espada et al 2004;Wu and Prior 2005), the 3-O-and 7-O-glucosides of the UV-protectant flavonols (Castillo-Muñoz et al 2007 and the 3-O/ 4 0 -O-glucosides of health beneficial stilbenes (Ali and Strommer 2003). Glucosylated anthocyanins, flavonols and stilbenes have also been identified in grape leaves and flowers (Langcake and Pryce 1977;Moore and Giannasi 1994;Schneider et al 2008).…”
