A simple model for the early events of quorum sensing in Pseudomonas aeruginosa: modeling bacterial swarming as the movement of an "activation zone"

Abstract: Background: Quorum sensing (QS) is a form of gene regulation based on cell-density that depends on inter-cellular communication. While there are a variety of models for bacterial colony morphology, there is little work linking QS genes to movement in an open system.

“…Another important result of their work is that the speed of the travelling wave of QS activation increases with the diffusion rate of the QS signal and decreases with the decay rate of the QS signal. Netotea et al (2009) presented an 2D agent-based computational model for the onset of swarming in P. aeruginosa, which is controlled by threshold levels of AHL signals and secreted factors (public goods). Their experimental framework utilizes swarming agar plates that allow the growth of activated bacteria, but not of non-activated ones.…”

“…We share the widespread opinion that the "change of bacterial lifestyle" is crucial for colonizing habitats and infecting susceptible hosts -unfortunately the signalling systems that orchestrate the underlying communication and collaboration mechanisms are not accurately annotated in bacterial genomes. Therefore, a systematic characterization of QS systems in Gram negative bacteria was carried out [68,69] and a modelling effort to map out the theoretically possible consequences of communication and collaboration in bacterial populations was initiated [70][71][72]. Virulence and adaptability of many Gram-negative bacterial species are associated with an N-acylhomoserine lactone (AHL) gene regulation mechanism called quorum sensing (QS).…”

“…A quorum érzékelés hatékony modellezéséhez azonban szükség van újabb, a sejtek által termelt kémiai komponensek, a kommunikációs jel és a közjavak reprezentációjára, melyek szabadon diffundálnak a környezetben. Így munkánk során egy olyan hibrid ágens modellt fejlesztettünk ki, mely egy kvalitatív szempontból helyes quorum érzékelési modellnek tekinthető [107]. A modell részletes bemutatására az Módszerek 3.4.…”

“…A Netotea-féle hibrid rendszer [107] első verziója Java programozási nyelven íródott, melyet a későbbiekben MATLAB programozási környezetben fejlesztettünk tovább. A két verzió ugyanazon algoritmus alapján működik, viszont a MATLAB verzió tervezése során kiemelt figyelmet fordítottunk a forráskód optimalizációjára, mely által a szimulációk is gyorsabbak lett.…”
