A simple technique for long‐term preservation of leptospires

Abstract: The viability of six serovars of Leptospira spp. was studied after long storage at −70°C. The bacteria were either preserved in Ellinghausen–McCullough–Johnson–Harris (EMJH) liquid growth medium or in sheep blood added as a cryoprotectant. The viability of the strains was observed on a monthly basis by dark‐ground microscopy over a period of 20 months at −70°C. Addition of sheep blood was not significantly advantageous, since leptospires that were stored in EMJH showed a slight increase in number after recover… Show more

“…As the virulence of pathogenic Leptospira tends to attenuate during in vitro passages and freeze storage (Reed et al., 2000; Samir and Wasfy, 2013), the assessment of virulence requires a restoration step in hamsters by serial infections (Haake, 2006). For this, two serial passages in hamsters were performed and the resulting isolates were subsequently used for experimental infection.…”

“…T h e r e s e a r c h e r c l a s s i f i e d t h e Cryoprotectants as penetrating or non-penetrating the cell, Glycerol belongs to the latter one 3 . It is one of the most important types of cell-conserving substances 18 , that act to surround the cells, delay the freezing of intracellular substances in these cells and reduce the solution effects 1 . It prevents ice crystallization and as a result, bacteria remain alive longer 3 , it is a strong substance that prevents water from entering the cell 19 .…”
