A small protein encoded by a putative lncRNA regulates apoptosis and tumorigenicity in human colorectal cancer cells

Abstract: Long noncoding RNAs (lncRNAs) are often associated with polysomes, indicating coding potential. However, only a handful of endogenous proteins encoded by putative lncRNAs have been identified and assigned a function. Here, we report the discovery of a putative gastrointestinal tract-specific lncRNA (LINC00675) that is regulated by the pioneer transcription factor FOXA1 and encodes a conserved small protein of 79 amino acids which we termed FORCP (FOXA1-Regulated Conserved Small Protein). FORCP transcript is un… Show more

“…FORCP is an ER-localized protein with two predicted transmembrane domains. Upon ER stress, FORCP inhibits basal proliferation and induces apoptosis [104]. However, the molecular mechanisms by which PIGBOS or FORCP regulates apoptosis in response to stress are unknown.…”

“…The FORCP transcript was not detected in most cell types, but was abundant in well-differentiated colorectal cancer cells, suggesting that FORCP is a novel small conserved protein encoded by misannotated lncRNA. FORCP can inhibit cell proliferation, clone formation and tumorigenesis ( 69 ).…”

“…LncRNAs play various roles in biological processes, including splicing, 48 protein localization, 49 and cellular proliferation. 50,51 LncRNAs are highly cell type specific in their expression patterns which makes them attractive as disease biomarkers for. 52 This is something that could be a particularly attractive area of investigation for autoinflammatory conditions such as arthritis and SLE which are notoriously difficult to diagnose quickly in the clinic.…”
