E. coli is one of the major significant pathogens causing mastitis, the most complex and costly diseases in the dairy industry worldwide. Present study was undertaken to isolate, detect the virulence factors, phylogroup, antimicrobial susceptibility and antimicrobial resistance genes in E. coli from cows with clinical mastitis. A total of 68 milk samples comprising 53 from clinical mastitis and 15 from apparently healthy cattle were collected from four different established dairy farms in Bangladesh. E. coli was isolated from the milk samples and identified by PCR targeting malB gene and sequencing of 16S rRNA gene. E. coli isolates were screened by PCR for the detection of major virulence genes ( stx , eae and cdt ) of diarrheagenic E. coli followed by phylogenetic grouping. Antimicrobial susceptibility of the E. coli isolates was determined by disk diffusion test and E. coli showing resistance was further screened for the presence of antimicrobial resistance genes. E. coli was isolated from 35.8% of the mastitis milk samples but none from the apparently healthy cattle milk. All the E. coli isolates were negative for stx , eae and cdt genes and belonged to the phylogenetic groups A and B1 which comprising of commensal E. coli . Antibiotic sensitivity testing revealed 84.2% (16/19) of the isolates as multidrug resistant. Highest resistance was observed against amoxicillin (94.5%) followed by ampicillin (89.5%) and tetracycline (89.5%). E. coli were found resistant against all the classes of antimicrobials used at the farm level. Tetracycline resistance gene ( tetA ) was detected in 100% of the tetracycline resistant E. coli and bla TEM-1 was present in 38.9% of the E. coli isolates. Findings of this study indicate a potential threat of developing antimicrobial resistance in commensal E. coli and their association with clinical mastitis. Occurrence of multidrug resistant E. coli might be responsible for the failure of antibiotic therapies in clinical mastitis as well as pose potential threat of transmitting and development of antibiotic resistance in human.

Subclinical mastitis (SCM) in water buffalo is a production disease associated with decreased milk yield and impaired milk quality and safety. Water buffalo is an important livestock species in Bangladesh, but information about the occurrence and aetiology of SCM in this species is scarce. A cross-sectional study was conducted as part of the Udder Health Bangladesh Programme to (i) determine the occurrence of SCM and bulk milk somatic cell count (SCC) in water buffalo in Bangladesh, (ii) identify pathogens causing SCM and (iii) evaluate penicillin resistance in isolated staphylococci strains. Sixteen buffalo farms in the Bagerhat and Noakhali regions of Bangladesh were selected for study and a bulk milk sample was collected from each farm. In addition, 299 udder quarter milk samples were collected from 76 animals. The bulk milk samples were assessed by direct SCC and the quarter milk samples by California mastitis test (CMT). The occurrence of SCM calculated at quarter and animal level was 42.5 and 81.6%, respectively. Milk samples from 108 CMT-positive quarters in 48 animals and 38 randomly selected CMT-negative quarters in 24 animals were investigated using bacteriological culture. Estimated mean bulk milk SCC was 195 000 cells/ml milk (range 47 000– 587 000 cells/ml milk). On culture, estimated quarter-level intramammary infection (IMI) was 40.4%. The identity of isolated bacteria was confirmed by MALDI-TOF mass spectrometry. Non-aureus staphylococci (NAS) were the most common pathogens (24.7%) and, among 36 NAS tested, 36.1% were resistant to penicillin. Thus there was high occurrence of SCM on the study farms, with relatively high penicillin resistance in NAS. Further studies are needed to identify underlying risk factors and develop an udder health control strategy for water buffalo in Bangladesh.

Mastitis is one of the diseases that cause economic losses worldwide due to the reduction in milk yield and the high treatment costs in dairy buffaloes. Although antibiotics are the mainstay treatment for this disease, the overuse of antibiotics has resulted in the emergence of antimicrobial resistance in animals and humans. Hence, this study aims to review and assess the available literature on bubaline mastitis in Asia. The prevalence of subclinical mastitis was higher in dairy buffaloes than in clinical mastitis, especially in Pakistan. Bubaline mastitis was commonly detected using the California mastitis test, surf field mastitis test, somatic cell count, and bacterial culture. In Asia, farm management and host factors were the primary causes of bubaline mastitis risk factors. Mastitis in buffaloes caused alterations in milk composition, such as increasing lactose levels, somatic cell count, and the presence of bacteria in the milk. However, protein, fat, and solid non-fat level variations were also affected by other factors such as the stage of lactation, breed, and age. The most prevalent isolated bacteria in bubaline mastitis milk samples were coagulase-negative staphylococci, Staphylococcus aureus, Streptococcus spp., and Escherichia coli. Most showed high resistance toward penicillin, amoxicillin, ampicillin, and streptomycin. The antimicrobial susceptibility of causative agents in Asia varies depending on the usage of common antibiotics to treat bubaline mastitis in each country. This review will help to understand bubaline mastitis better, although studies are limited in many Asian countries.
