A tetrodotoxin-resistant voltage-gated sodium channel expressed by sensory neurons

Abstract: Dorsal root ganglion sensory neurons associated with C-fibres, many of which are activated by tissue-damage, express an unusual voltage-gated sodium channel that is resistant to tetrodotoxin. We report here that we have identified a 1,957 amino-acid sodium channel in these cells that shows 65% identity with the rat cardiac tetrodotoxin-insensitive sodium channel, and is not expressed in other peripheral and central neurons, glia or non-neuronal tissues. In situ hybridization shows that the channel is expressed… Show more

“…Construction of cRNA for oocyte expression cDNAs for the human a3, a5, and b4 (GenBank accession numbers Y08418, Y08419 and Y08416, respectively), containing only coding sequences and an added Kozak consensus sequence (GCCACC) immediately upstream of the start codon (Groot-Kormelink & Luyten, 1997), were subcloned into the pSP64GL vector, which contains 5' and 3' untranslated Xenopus b-globin regions (Akopian et al, 1996). The mutants in 9' (a3 L279T , a5 V290L , a5 V290T and b4 L272T , where V, L and T stand for valine, leucine and threonine, respectively) were created using the QuickChange TM Site-Directed Mutagenesis Kit (Stratagene) and their full-length sequence was veri®ed.…”

“…Both scn5Laa and scn5Lab were also expressed in nervous and cardiac tissues. In mammals, SCN5A is expressed in cardiac muscle and limited regions of the brain (Hartmann et al 1999;Rogart et al 1989), while SCN10A and SCN11A are expressed in peripheral nociceptive sensory neurons (Akopian et al 1996;Dib-Hajj et al 1999a;Sangameswaran et al 1996). However, mammalian expression of SCN5A, SCN10A or SCN11A has not been reported in spinal cord or eye.…”
