A type II-collagen derived peptide and its nitrated form as new markers of inflammation and cartilage degradation in equine osteochondral lesions

Abstract: Markers of cartilage breakdown enable studying the degradation of cartilage matrix in equine joint pathologies. This study was designed to determine the levels of Coll2-1, a peptide of the triple helix of type II collagen, and Coll2-1NO 2 , its nitrated form in the plasma of healthy horses (controls; n = 37) and horses suffering from osteochondrosis (n = 34). Clinical and arthroscopic scores were attributed reflecting the severity of lesions and were related to the plasma levels of Coll2-1 and Coll2-1NO 2 . Th… Show more

“…This work adds important data regarding their qualifications. This study positioned Coll2-1 and Coll2-1 NO 2 biomarkers in the framework of biomarkers applied to the development of drugs intended for the treatment of OA as described in the recent published work by Kraus et al 35 Previous works reporting Coll2-1 and Coll2-1 NO 2 levels have fulfilled the step of exploration based on pre-clinical evidence [18][19][20][21][22] and the further step of demonstration giving evidence that support the primary clinical evidence in both healthy and OA patients. 16,17,23 The Coll2-1 AND Coll2-1 NO 2 LEVELS AFTER VISCOSUPPLEMENTATION 905 present study gave the first evidence to the next step of characterization.…”

“…15 Specific immunoassays have been developed to measure a specific peptide located in the triple helix, Coll2-1 and its nitrated form Coll2-1 NO 2 . 16,17 These biomarkers have been validated in vivo in mouse, guinea pig and horse [18][19][20][21][22] and also in healthy human 16 and OA patients. 17,23 The aim of this study was to measure the evolution of the serum levels of Coll2-1 and Coll2-1 NO 2 in knee OA patients after VS.…”

“…Thus, collagen is specific for cartilage, and investigation of its fragments can be taken as a marker of cartilage degeneration. Alterations of the major constituent of extracellular cartilage matrix, type II collagen, have also been noted [4].…”

“…Developmental osteochondral lesions are characterized by the presence of localized bone and cartilage defects and have been associated with markers of collagen degradation and inflammation (Billinghurst et al 2004;Gangl et al 2007), reflecting the presence of these cartilage lesions. Different studies suggest a role for IGF-I in restoring cartilage homeostasis following injury (Fortier et al 2001;Rogachefsky et al 1993;Semevolos et al 2001;Tuncel et al 2005).…”
