A useful monoclonal antibody (BL2-10D1) to identify tumor cells in urine cytology

Abstract: The monoclonal antibody (MoAb) BL2-10D1 directed against a tumor-associated antigen of human bladder cancer was used to identify tumor cells obtained by bladder washing or voided urine. The reactivity of BL2-10D1 MoAb was detected by an immunoperoxidase method and evaluated in ten healthy donors and in a series of 65 patients. The 65 patients studied were divided into three groups: ten with nontumor bladder disease (group A); 36 with bladder carcinoma (group B); and 19 with a history of bladder neoplasia but n… Show more

“…These include cytokeratin 18 (Ramaekers et al, 1985), Lewis X antigen (Cordon-Cardo et al, 1988;Sheinfeld et al, 1990), and 19A211-reactive cell-surface antigen (Fradet et al, 1990). The MAbs 486P3/12 (Huland et al, 1987), BL2-1OD1 (Longin et al, 1990), and IBE12 (Pancino et al, 1991) showed the same staining pattern, but all were suggested to be reactive with Lewis X antigen (Fradet et al, 1990;Pancino et al, 1991).…”

“…Positive staining occurred in 0%, 5% and none of the specimens from 15 patients without malignancies, 19 subjects with other urological malignancies, and 23 individuals showing a history of TCC but no detectable tumor, respectively. Antibody BL2-10D1 has been evaluated on bladder washings or voided urine of 65 patients [15]. After centrifugation of urine, the pellet was suspended in PBS and spun onto slides.…”

“…10 Recently, IM studies have shown that cells of well-and moderately differentiated TCCs reacted with monoclonal antibody BL2-10D1, while about 70% of poorly differentiated or grade III TCCs did not express any immunoreactivity with this antibody. 23,24 There are two other monoclonal antibodies to 486P 3/12 25,26 and CA 19-9 27 that have value in the identification of TCC cells. Of these two antibodies, 486 P 3/12 is expressed by about 90% of all-grade TCCs.…”
