Aberrant Notch signaling in glioblastoma stem cells contributes to tumor recurrence and invasion

Abstract: Abstract. Upregulation of the Notch signaling pathway in cancer stem cells and side population (SP) cells has a major role in maintenance, self-renewal and chemoresistance. The present study isolated a cancer stem cell-like SP accounting for 4.1% of a glioblastoma cell population using a Hoechst 33342 dye exclusion assay. In this glioblastoma SP, the expression of of Notch1 signaling proteins Notch1 intracellular domain and Hes-1 was markedly upregulated. Furthermore, knockdown of Notch1 by RNA interference si… Show more

“…PDGFR and NOTCH1 are highly enriched in GBM, and targeting these two genes to treat GBM is currently under investigation [2437]. By using the GlioVis program, we analyzed the mRNA levels of NOTCH1 gene and PDGFR isoforms (PDGFRA and PDGFRB) in more than 500 GBM patients from The Cancer Genome Atlas (TCGA) database.…”

“…Among these therapeutic targets, NOTCH1 [2431] and PDGFR [3237] (platelet-derived growth factor receptors) have received more attention recently given their important roles in GSCs. Our previous research demonstrates that NOTCH1-positive GSCs are more potent to self renew than NOTCH1-negative cells, suggesting that NOTCH1-positive GSCs are more tumorigenic [31].…”

“…In the case of solid tumours, expression of embryonic stem cell markers such as Oct4 and Sox2 is commonly seen in association with aberrant Notch expression [44] , it and may promote metastasis by modulating epithelialto-mesenchymal transition programmes, a process also known to involve Hedgehog signalling. Future work will hopefully pave the way for the development of new strategies for targeting these pervasive oncogenic pathways.…”

“…Although the role of stem-like cells has been extensively evaluated, their contribution to relapse, chemo- and radio-therapy resistance [2] and the role of vascular cells such as microglia, peripheral immune and neural processor cells; in the generation of a specific niche within which GBM cells can evade immune detection is a topic of ongoing research. A detailed understanding of the supportive role that the microenvironment plays in GBM is critical to the design of effective immunotherapeutic strategies.…”
