Absorption Spectra of Planarian Visual Pigments and Two States of the Metarhodopsin Intermediates

Abstract: Abstract— Absorption spectra measurements of isolated planarian ocelli by a microspectrophotometer (MSP) and intra‐ocellar recordings of the early receptor potential (ERP) were carried out in order to characterize in situ planarian rhodopsin (pRh) and its photoproducts. The MSP spectra of the isolated ocelli revealed Λmax at about 500 nm. The ERP evoked by a test stimulus was a positive monophasic waveform in the dark but became negative during exposure to violet light. During subsequent darkness, the ERP rap… Show more

“…The behavior trial lasted 18 h and consisted of the following settings. Background illumination in all quadrants was 50 lm red light, which is undetectable to planarians given their photoreceptor profiles (Azuma, Iwasaki, & Ohtsu, ; Brown & Ogden, ). In addition, half of the arena is illuminated by 420 lm blue light, giving the animals a choice between “light” and “dark” halves of the environment.…”

“…The trajectory of movement was analyzed with SMART software (Panlab) (Figure 1 C) [ 6 ]. All behavioral experiments were performed in a dark room with only a red light of a wavelength that cannot be sensed by planarians (Figure 1 C) [ 18 ]. Calculations based on the data obtained were performed using static ggplot2 package of R software [ 19 ].…”

“…The pigment cells are arranged into a semilunar eyecup, while the visual bipolar neurons are located outside of the eyecup. Dendrites from the visual neurons distribute inside of the pigment eyecup to form a rhabdomeric structure containing opsins (Asano et al ., 1998;Orii et al ., 1998;Azuma et al ., 1999;. The axons of the visual neurons project caudally onto the dorso-medial region of the brain and some of the fibers form an optic chiasma, which integrates photosensory inputs from the left and right sides of the animal (Agata et al ., 1998;Sakai et al ., 2000).…”
