Abundance and community structure of ammonia-oxidizing microorganisms in reservoir sediment and adjacent soils

Abstract: Ammonia oxidation is an important process for global nitrogen cycling. Both ammonia-oxidizing bacteria (AOB) and archaea (AOA) can be the important players in nitrification process. However, their relative contribution to nitrification remains controversial. This study investigated the abundance and community structure of AOA and AOB in sediment of Miyun Reservoir and adjacent soils. Quantitative PCR assays indicated that the highest AOA abundance occurred in unplanted riparian soil, followed by reservoir sedi… Show more

“…Most of the AOB amoA OTUs belongs to cluster three (63 OTUs, 61 %), which the most dominant cluster. The dominance of Nitrosospira was already reported in earlier studies showing that Nitrosospira was the majority species in soil ecosystems (Chen et al 2011;Xu et al 2012;Wang et al 2013) including riparian or agricultural soil sampled from the watershed of the Miyun reservoir. Furthermore, the dominance of Nitrosospira were also verified in Cu-or Zn-contaminated soil by Mertens et al (2009) andLiu et al (2014).…”

“…Microorganisms from archaeal class Thaumarchaeota can play an important role in ammonia oxidation in the environment (Jung et al 2014;Wang et al 2014). So far, the dominance of Thaumarchaeota in lake sediment archaeal community has rarely been reported.…”

“…The specific primer sets Arch-amoAF/Arch-amoAR and AmoA-1F/ AmoA-2R were applied for quantitative PCR (qPCR) assay of the archaeal and bacterial amoA genes, and construction of AOA and AOB clone libraries, following the previously reported conditions Wang et al 2014a). One-way analysis of variance (ANOVA) followed by Student-Newman-Keuls test was used to check the quantitative differences (P<0.05) in the abundance of AOA or AOB amoA gene among different soils.…”

“…such as moisture (Vasileiadis et al 2012), pH (Nicol et al 2008), carbon-and nitrogen-related fertility (Wang et al 2014c;Wessen et al 2010;Wertz et al 2012), climate (Yao et al 2013), and pesticide (Wan et al 2014a, b). Moreover, it has been well-documented that land use type can also affect the distribution of soil ammonia-oxidizing microorganisms Meyer et al 2014;Qin et al 2013;Wang et al 2014a;Zhao et al 2015). Multiple environmental factors may collectively regulate the distribution of AOA and AOB in soil ecosystems (Jiang et al 2014;Yao et al 2013), and no single factor can discriminate between AOA and AOB (Prosser and Nicol 2012).…”
