Four 3,5-pyrazolidinedione derivatives namely, 4-(4`-chlorobenzylidene)-1-phenylpyrazolidine-3,5-dione (1), 4-(4`-nitrobenzylidene)-1-phenylpyrazolidine-3,5-dione (2), 2'-(4-chlorophenyl)-1phenyl-3,5-dioxo-1,2,3,5 tetrahydrospiropyrazole-4,3' oxirane (3) and 2'-(4-nitrophenyl)-1phenyl-3,5-dioxo-1,2,3,5 tetrahydrospiropyrazole-4,3' oxirane (4) were prepared in pure state and bioassayed against 2 nd and 4 th instars larvae of cotton leaf worm, Spodoptera littoralis (Boised) (Lepidoptera: Noctuidae) using feeding and dipping bioassay. The results of bioassays indicated that title compounds exhibit satisfactory insecticidal activities. Among those, compound (1) exhibit the highest insecticidal activities against 2 nd instar larvae, with LC 50s 3.23 and 0.619 mgL -1 for feeding, and 36.04 and 28.69 mgL -1 for dipping, after 48 and 72 h treatment. According to the toxicity index the compound (1) showed the highest larvicidal activity against 4 th instar larvae with LC 50s 141.33 and 76.12 mgL -1 for feeding larvae, and 26.94 and 12.29 mgL -1 for dipping larvae after 48 and 72 h treatment. These results showed that, the 2 nd larvae was more susceptible than 4 th instars larvae to these compounds. In addition, the insecticidal activity of these compounds was Aly A. Abd-Ella et al. 2 more effective on cotton leaf worm larvae in feeding bioassay as compared with the dipping treatment. The rest of the tested compounds possessed moderate to strong larvicidal activities against cotton leaf worm. In general, the results indicate the possible use of 3,5-pyrazolidinedione derivatives as components in integrated pest management program against S. littoralis.

Bioluminescence in the ophiuroid Amphipholis squamata is produced by photocytes located within the spinal ganglia of arm spines. Ganglionic cells were dissociated (pronase digestion) and photocytes separated from other cell types by using a continuous density Percoll gradient. Aliquots from a stock suspension of photocytes in artificial sea water were stimulated to produce light by using KCl or acetylcholine and fixed for ultrastructural observation at different times of the luminous process. Preluminescent, luminescent, and postluminescent photocytes contained various intracytoplasmic structures, such as Golgi, flat and distended rough endoplasmic reticulum cisternae, bundles of fibrils, and up to six types of membrane-bounded vesicles. These structures either co-occurred or succeeded one another during the process of light production, indicating that they were most probably participating in the luminescence reaction. Two types of vesicles, sharing some ultrastructural features, probably represented the microsources of the photocytes. One type occurred almost exclusively in luminescent photocytes, and the other almost exclusively in postluminescent photocytes, suggesting that one may be transformed into the other. The latter type of vesicle contained densely packed fibro-tubular units, giving a characteristic paracrystalline appearance to postluminescent photocytes.

Piperidine and cyclohexyl ring homologues of the high-affinity dopamine (DA) uptake inhibitor 1-[1-(2-benzo[b]thienyl)cyclohexyl]piperidine (BTCP, 3) were each prepared in four steps from the appropriate cycloalkanones. These compounds were tested for their ability to displace [3H]BTCP and [3H]cocaine and to inhibit [3H]DA uptake in rat striatal homogenates. The ratios IC50([3H]cocaine)/IC50([3H]BTCP) ranged from 62 for BTCP to 1.5 for 1-[2-(benzo[b]thienyl)-cyclopentylamine (17); cocaine gave a ratio of 0.6. This indicates that BTCP is the most selective of all the compounds tested for sites labeled by [3H]BTCP whereas cocaine is most selective for sites labeled by [3H]cocaine. The wide differences in the relative abilities of these compounds to displace [3H]BTCP and [3H]cocaine suggests that these two radioligands are labeling different sites on the transporter. In general, the compounds structurally related to BTCP exhibited greater selectivity for sites labeled by [3H]BTCP. However, several of the BTCP-related derivatives showed greater (compared with BTCP and cocaine) ability to displace [3H]cocaine. Most notably, 1-[1-(2-benzo[b]thienyl)cyclohexyl]pyrrolidine (7) exhibited a 3.4-fold greater affinity for these sites compared with BTCP and a 9-fold greater affinity at these sites than cocaine. Most of the BTCP homologues displayed greater ability to inhibit [3H]DA uptake in rat forebrain synaptosomes than cocaine. BTCP and 7 were the most potent of all the compounds tested in terms of their ability to inhibit uptake of [3H]DA. IC50 ratios for [3H]cocaine binding/[3H]DA uptake ranged from 0.47 for 1-[1-(2-benzo[b]thienyl)cyclopentyl]homopiperidine (11) to 8.8 for 1-(2-benzo[b]thienyl)cyclohexylamine (4). The importance of this ratio remains unclear in terms of identification of potential cocaine antagonists. As for BTCP, all of the compounds tested showed Ki values > 10,000 nM for displacement of [3H]TCP from rat brain homogenates. These compounds were able to displace the highly selective sigma receptor probe [3H]-(+)-pentazocine from guinea pig brain homogenates with Ki values ranging from 125 to 9170 nM. The significance of their sigma-binding activity in light of their dopaminergic properties is unclear. The diverse binding properties of these compounds at the DA-uptake site and their spectrum of inhibitory activities for [3H]DA uptake identifies them as a useful base for the development of subtype selective probes at this site. These compounds will allow further study of the structure and function of the "cocaine" receptor as well as the development of potential cocaine antagonists.
