Activation of NLRC4 downregulates TLR5-mediated antibody immune responses against flagellin

Abstract: Bacterial flagellin is a unique pathogen-associated molecular pattern (PAMP), which can be recognized by surface localized Toll-like receptor 5 (TLR5) and the cytosolic NOD-like receptor (NLR) protein 4 (NLRC4) receptors. Serial dilutions of serum or saliva samples were added in each well and incubated at 37 C for 2 h. After 6washes with PBS containing 0.5% Tween-20 (PBST), the plates were incubated with 100 ml of alkaline phosphatase-conjugated goat antimouse/rat heavy chain IgG or IgA at 37 C for 1 h. After 6 washes of PBST, 100 ml phosphate substrate (p-nitrophenylphosphate) was added into each well for color development. Endpoint titers were expressed as the reciprocal of the last dilution giving an optical density at 405 nm of 0.1 greater than that of control after 30 min of incubation.…”
