Activation of the p38 and p42/p44 mitogen‐activated protein kinase families by the histamine H1 receptor in DDT1MF‐2 cells

Abstract: 1 The mitogen-activated protein kinases (MAPKs) consist of the p42/p44 MAPKs and the stressactivated protein kinases, c-Jun N-terminal kinase (JNK) and p38 MAPK. “…These observations re¯ect the accepted coupling of the histamine H 1 receptor to phospholipase C activation via G q/11 -proteins (Hill et al, 1997). However, we have recently shown that histamine H 1 receptor-induced increases in p42/ p44 MAPK and p38 MAPK in DDT 1 MF-2 cells were partially sensitive to pertussis toxin suggesting coupling to members of the G i /G o -protein family (Robinson & Dickenson, 2001). Hence, in this study the role of G i /G o -proteins in histamine H 1 receptor-induced phosphorylation of PKB was determined in cells exposed to pertussis toxin (100 ng ml 71 ) for 16 h. As shown in Figure 3b, pertussis toxin pre-treatment virtually abolished histamine-induced increases in PKB phosphorylation.…”
