Activation tagging in indica rice identifies ribosomal proteins as potential targets for manipulation of water‐use efficiency and abiotic stress tolerance in plants

Abstract: We have generated 3900 enhancer-based activation-tagged plants, in addition to 1030 stable Dissociator-enhancer plants in a widely cultivated indica rice variety, BPT-5204. Of them, 3000 were screened for water-use efficiency (WUE) by analysing photosynthetic quantum efficiency and yield-related attributes under water-limiting conditions that identified 200 activation-tagged mutants, which were analysed for flanking sequences at the site of enhancer integration in the genome. We have further selected five plan… Show more

“…RPL6 and RPL23A, in addition to their role in WUE, became up-regulated in almost all the stresses studied illustrating their possible involvement in inducing tolerance to abiotic stresses (Moin et al, 2016). Cold, MeJa, and SA treatments induced the up-regulation of a majority of RPL genes, while H 2 O 2 and heat treatments down-regulated 75% of the genes.…”

“…TOR is a highly conserved master coordinator of nutrients, energy and stress signaling in the cell (Xiong and Sheen, 2014). Thus, implying a role of RPs in the direct response to stress, through a specific network of genes related to stress in plants, such as defense proteins, ROS response genes, genes of calcium signaling and the vast network of genes associated with phytohormones (Moin et al, 2016). Finally, given the induction of RPS6 in the vanilla transcriptome in response to Fusarium , this may suggest the participation of TOR master regulator in the translational regulation in the Vanilla- Fusarium pathosystem.…”

1The extra-ribosomal functions of ribosomal proteins RPL6 and RPL23a in stress-2 responsiveness have emanated from our previous studies on activation tagged mutants of rice 3 screened for water-use efficiency (Moin et al, 2016a). In the present study, we functionally 4 validated the RPL6, a Ribosomal Protein Large subunit member for salt stress tolerance in 5 rice.
