Active matrix metalloproteinase-8 (aMMP-8) point-of-care test (POCT) in the COVID-19 pandemic

Abstract: Introduction Active matrix metalloproteinase (aMMP)-8 utilized in point-of-care testing (POCT) is regarded as a potential biomarker for periodontal and peri-implant diseases. Various host and microbial factors eventually influence the expression, degranulation, levels and activation of aMMP-8. The type of oral fluids (saliva, mouthrinse, gingival crevicular, and peri-implant sulcular fluids [GCF/PISF], respectively) affect the analysis. Areas covered With this backgroun… Show more

“…Since the start of the pandemic in 2019, our team has been working diligently towards unravelling the intricacies of this association [ 2 6 ]. An association between periodontitis and higher risk of Intensive Care Unit admission, need for ventilation and death of COVID-19 patients, is now well documented [ 3 ].…”

“…Hence any diagnostic modality which could predictably screen the presence of periodontal disease could very well act as an alarm to encourage and motivate the patients to seek dental care before it has progressed to severe stage. Recently developed and validated active matrix metalloproteinase (aMMP)-8 point-of-care/chair-side biomarker technology provides possibilities for health care professionals in this regard [ 6 ]. These lateral flow immunoassay based kits are capable of diagnosing active periodontal disease at both full-mouth and site-specific levels and can aid in tailoring therapy as per patient requirements and at the same time sensitizing them towards the need to maintain oral health [ 6 ].…”

“…According to Sorsa et al, the active metalloproteinase-8 point-of-care test (aMMP-8 POCT) provides a simple, Non-invasive, rapid, and real-time tool that may be used to identify the Peri-implant disease as a potential risk factor of COVID-19 [ 63 ]. The aMMP-8 levels above 20 ng/ml are indicative of active Peri-implant disease.…”

“…The visible result on the dipstick was documented with a photograph too. Among the positive cases with two lines there existed both weak or thin(ner) (a weak positive) or strong or thick(er) (a strong positive) second line (Sorsa et al, 2021 ). The remaining elution fluid of PISF was used for further analysis in the study and was analyzed for MMP‐2 and MMP‐9 as well as total MMP‐8, IL‐6, and calprotectin by Western immunoblotting, gelatin‐zymography, and ELISA as described in Sections 2.2.2 , 2.2.3 , 2.2.4 .…”
